Cytokine Priming

Cytokine priming is a biological process in which prior exposure to signaling proteins called cytokines alters a cell’s response to a later stimulus, helping regulate the strength and timing of immunity. During priming, cytokine-receptor signaling activates transcription factors and can change gene expression, cellular metabolism, or antimicrobial functions, so subsequent recognition of a pathogen produces an enhanced or more rapid response. In immunology and infection research, this process is studied in macrophages, neutrophils, and other immune cells to understand host defense, inflammatory injury, and pathogen control. Its mechanisms also inform vaccine development and strategies for modulating immune responses.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This technique provides a method to harvest, normalize and quantify intracellular growth of bacterial pathogens that are pre-cultivated in natural protozoan host cells prior to infections of mammalian cells.
