Intestinal Tissue Preparation

Intestinal tissue preparation is a set of laboratory methods used to collect, preserve, process, and section intestinal samples for microscopic examination, making tissue architecture and cellular changes suitable for medical analysis. Typically, specimens are trimmed and fixed to stabilize cellular structures, then dehydrated, embedded in a supporting medium, sectioned into thin slices, and stained to reveal features such as mucosa, glands, connective tissue, and inflammatory or neoplastic changes. These preparations support routine histopathology, immunohistochemistry, and research on intestinal disease, including infection, inflammation, malabsorption, and cancer. Consistent handling improves tissue quality, diagnostic interpretation, and reproducibility across clinical and experimental studies.

Accurate identification and location of epithelial cells along the intestinal mucosal lining are essential to define different cell lineages. Proper imaging of intestinal tissues is crucial for identification of protein expression patterns with maximum resolution. This study aims to delineate the optimal methods and conditions for processing mouse intestinal tissues.

Recently available video recording and spatiotemporal mapping (STmap) techniques make it possible to visualize and quantify both propagating and mixing patterns of intestinal motility. The goal of this protocol is to explain the generation and analysis of STmaps using the GastroIntestinal Motility Monitoring (GIMM) system.
