Needle Knife Therapy

Needle knife therapy, also called acupotomy, is a minimally invasive treatment that combines a needle-like instrument with a small blade to address localized soft-tissue disorders and pain. During the procedure, a clinician guides the instrument into targeted tissue and uses controlled cutting or release motions to disrupt adhesions, fibrotic bands, or contracted structures that may restrict movement. In medicine, the technique is applied primarily in musculoskeletal care, including selected cases of chronic pain, stiffness, and impaired function. Its potential to modify abnormal tissue tension while limiting the size of an incision makes it a subject of clinical research and procedural training.

We describe here a protocol for microinjection in the rodent brain that uses quartz needles. These needles do not produce detectable tissue damage and ensure reliable delivery even in deep regions. Moreover, they can be adapted to research needs by personalized designs and can be re-used.
