Alpha Receptor Selectivity

Alpha receptor selectivity is the preferential interaction of a drug with one adrenergic receptor subtype over others, an important principle for predicting pharmacological effects and adverse reactions. In pharmacology, selectivity reflects differences in drug affinity and receptor distribution, with alpha-1 receptors primarily activating Gq-mediated phospholipase C signaling and alpha-2 receptors coupling to Gi to reduce cyclic AMP and inhibit neurotransmitter release. Selective agonists and antagonists can therefore produce targeted changes in vascular tone, blood pressure, sympathetic signaling, or neurotransmission. Understanding these relationships supports drug design, therapeutic selection, dose optimization, and interpretation of responses that may change as drug concentrations increase.

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.

Peripheral injection of alpha-synuclein fibrils into the peritoneum or tongue of Tg(M83+/-:Gfap-luc+/-) mice, which express human alpha-synuclein with the familial A53T mutation and firefly luciferase, can induce neuropathology, including neuroinflammation, in their central nervous system.
