1. Voorbereiding

Voordat u begint, draag laboratoriumhandschoenen en de juiste beschermende kleding.
Steriliseer alle dissectie-instrumenten, eerst met een reinigingsmiddel en vervolgens met 70% ethanol en veeg ze dan grondig droog.
Bereid 50 mL Hank's gebalanceerde zoutoplossing (HBSS) met 2% foetaal runder serum (FCS) voor.

2. Dissectoe

Gebruik een kooldioxide-afleversysteem om de muis te euthanaseren door middel van hypoxie. Bevestig de geëuthaniseerde muis in rugligging op een dissectieplaat en voer een longitudinale laparotomie uit met behulp van een schaar en pincetten.
Gebruik pincetten om de darmen en maag aan de rechterkant van de buik te verplaatsen om de maag en milt bloot te leggen. De milt is aan de maag gehecht.
Gebruik pincetten om de milt voorzichtig van de maag los te maken en plaats deze in de petrischaal met 5 mL HBSS 2% FCS.

3. Immuuncelisolatie

Plaats de milt op een 40 µm celfilter boven dezelfde petrischaal. Verpletter de milt met een plunjer om het te dissociëren.
Breng de gedissocieerde milt en de vloeistof over in een 15 mL centrifugebuis.
Centrifugeer de buis gedurende 7 min bij 370 x g en 10°C en gooi het supernatant weg terwijl het pellet wordt behouden.
Resuspendeer het pellet in 2 mL kaliumacetaat om de erytrocyten te lyseren. Wacht 2 min en breng het volume vervolgens op 15 mL met HBSS 2% FCS.
Centrifugeer de buis opnieuw gedurende 7 min bij 370 x g en 10°C. Gooi het supernatant weg en resuspendeer het pellet in 5 mL HBSS 2% FCS.
Tel de cellen met behulp van een trypaanblauwkleuringstest en pas de uiteindelijke celconcentratie aan op 10⁷ cellen/mL met behulp van een geschikt volume HBSS 2% FCS.

4. CFSE-kleuring en T-celstimulatie

Verdeel 10⁷ geïsoleerde miltcellen/buis in 4 buizen (15 mL buizen, gelabeld 1 tot 4)
Voeg 3 mL HBSS 2%FCS toe aan elke buis.
Voeg 1 µL CFSE toe aan elke buis (eindconcentratie - 5 µM).
Incubeer de buizen gedurende 10 min bij 37°C in een 5% CO₂ incubator.
Voeg in buis 3 en 4 12 mL HBSS 2% FCS + anti-CD3 antilichaam toe met een eindconcentratie van 2,5 µg/mL. Buis 3 en 4 worden gestimuleerd met anti-CD3 antilichaam om het effect op de celcyclus te observeren.
Voeg in buis 1 en 2 12 mL HBSS 2% FCS toe. De cellen in buis 1 en 2 worden niet gestimuleerd.
Centrifugeer alle buizen gedurende 7 min bij 370 x g en 10°C. Gooi de supernatanten weg.
Resuspendeer de pellets in 2 mL HBSS 2%FCS.
Breng de resulterende oplossingen over in afzonderlijke putjes op een 6-putjesplaat.
Incubeer de cellen gedurende 3 dagen bij 37°C, 5% CO₂.

5. Celkleuring

Op dag 3, voeg 2 mL HBSS 2%FCS toe aan put 1 en 3.
Pipet krachtig en breng de monsters over in 5 mL FACS-buizen.
Vervolg het incuberen van de resterende cellen uit putten 2-4 bij 37°C, 5% CO₂. Ze zullen worden geanalyseerd op dag 5 om de langetermijneffecten van stimulatie op de celcyclus te onderzoeken.
Centrifugeer de buizen gedurende 7 min bij 370 x g en 10°C. Gooi de supernatanten weg.
Voeg 100 µL antilichaammix (zie Tabel 1) toe aan elke buis.

Incubeer de buizen gedurende 20 min op ijs in het donker.
Voeg 1 mL HBSS 2%FCS toe en centrifugeer de buizen gedurende 7 min bij 370 x g en 10°C.
Gooi het supernatant weg en resuspendeer de pellets in 200 µL HBSS 2% FCS.
Breng de resuspendeerde pellets over in nieuwe, gelabelde FACS-buizen.
Evalueer de T-celproliferatie met behulp van FACS.
Herhaal op dag 5 het celkleuringsproces met de cellen uit de overige twee putjes van de 6-putjesplaat.

6. Gegevensanalyse

Open de "FlowJo" software en sleep de bestanden naar het "Alle monsters" venster.
Dubbelklik op het bestand voor niet-gestimuleerde cellen die op
