Question 1

Why are fruit flies and roundworms used to study aging and lifespan?

Accepted Answer

Invertebrate model organisms like Drosophila melanogaster and Caenorhabditis elegans have short generation times, allowing scientists to observe multiple generations over days or weeks rather than months. They are also highly amenable to genetic manipulation with tools that enable specific genes to be turned on or off easily, making them ideal for examining genetic and environmental influences on aging.

Question 2

What is the dauer stage and why is it important in worm lifespan experiments?

Accepted Answer

Dauer is a hardy, growth-arrested state that worm larvae enter when starved. To synchronize age-matched worms for lifespan studies, researchers induce dauer by starving worms on a culture plate for approximately one week. When transferred to food-rich environments, dauer larvae resume their life cycle and become reproductively mature, allowing scientists to obtain cohorts of uniformly aged animals for consistent experimental measurements.

Question 3

How does FUDR treatment support lifespan measurement in worms?

Accepted Answer

FUDR is a drug that suppresses worm reproduction without affecting adult lifespan, making it essential for accurate lifespan quantification. By preventing reproduction, FUDR eliminates confounding variables from offspring and allows researchers to focus solely on measuring the survival of individual worms over time without interference from progeny.

Question 4

What steps are involved in preparing age-matched fruit flies for lifespan studies?

Accepted Answer

Adult flies are placed in an egg collection cage with fruit juice-agar plates and yeast paste. Collected eggs are washed and incubated for about 10 days to develop into flies. One-day-old adults are transferred to food bottles for two days to reach sexual maturity, then anaesthetized and sorted by sex to avoid confounding lifespan measurements with sex-specific factors.

Question 5

How can RNAi be combined with lifespan assays to study aging?

Accepted Answer

RNA interference, or RNAi, uses short RNAs to decrease the expression level of target genes. Scientists can treat worms with RNAi against specific genes, then perform survival assays to observe how gene knockdown affects lifespan. This technique helps identify genes that influence animal survival and aging processes by comparing lifespan in knockdown organisms versus controls with normal gene expression.

Question 6

What environmental factors have been shown to influence invertebrate lifespan?

Accepted Answer

Temperature, drugs, pathogens, and diet significantly affect lifespan in invertebrate models. For example, growing worms at 25°C rather than 20°C decreased lifespan, as did high concentrations of the antidepressant Mirtazepine. Caloric or dietary restriction has also been observed to have significant effects on worm lifespan, demonstrating how growth conditions directly impact longevity.

Question 7

How are high-throughput lifespan experiments conducted in invertebrates?

Accepted Answer

Researchers culture worms in liquid media in 96-well microtiter plates to test multiple substances and growth conditions simultaneously. This approach allows scientists to screen the effects of various drugs, environmental factors, and genetic modifications on lifespan in parallel. The data generated helps identify pathways controlling aging and supports the development of therapies for age-related diseases like Alzheimer's and cardiovascular disease.
