Welkom bij de JoVE-methodecollecties over Chloroplast-onderzoeksmethoden, samengesteld door onze gasteditor, Paul Jarvis. Deze collectie behandelt een breed scala aan in vitro- en in vivo-technieken die eiwitinteracties binnen chloroplasten karakteriseren. Onderzoekers aan de Universiteit van Oxford beschrijven een in vitro-methode met behulp van geïsoleerde chloroplasten, die worden geïncubeerd met radioactief gelabelde eiwitten om hun opname te monitoren.
Deze methode is een ontwikkeling van eerdere benaderingen die werden gebruikt om chloroplast-eiwitimport in vitro te bestuderen. Vervolgens wordt in een aanvullende methode de chloroplast-eiwitimport in vivo onderzocht. Deze methode gebruikt tijdelijke expressie van chimere eiwitten en protoplasten om eiwitgrootteveranderingen en lokalisatie te onderzoeken via immunoblotting en fluorescentiemicroscopie.
In deze methodevideo onderzochten de auteurs het thylakoïd-membraansysteem aanwezig in chloroplasten. Door de membranen te isoleren, worden eiwittranslocatieroutes in chloroplasten geëlucideerd door SDS-PAGE en fosforbeeldvorming. Onderzoekers in Frankrijk passen verschillende centrifugatiemethoden toe om de subcompartimenten in chloroplasten te isoleren.
Deze geïsoleerde cellulaire fracties worden vervolgens geanalyseerd door immunoblotting en proteomics om informatie te verkrijgen over de sub-organellaire lokalisatie van chloroplast-eiwitten. Vervolgens gebruikten Zwitserse onderzoekers affiniteitszuivering van chloroplast-multi-eiwitcomplexen om de moleculaire machinerie betrokken bij eiwitimport te onderzoeken. Specifiek de talk en tick multi-eiwitcomplexen ingebed in envelopmembranen.
In een aanvullende benadering karakteriseren wetenschappers het plantaardige thylakoïd-eiwitcomplex in Arabidopsis met behulp van Blue Native PAGE, of BN-PAGE, die de scheiding van eiwitcomplexen in hun native en functionele vorm mogelijk maakt. De complexen worden verder geanalyseerd via 2D SDS-PAGE. This JoVE Methods Collection on Chloroplast Research Methods presents a comprehensive set of complementary in vitro and in vivo techniques for studying chloroplast protein import, localization, suborganellar fractionation, and protein-protein interactions. The collection focuses on model plant Arabidopsis thaliana but is adaptable to other organisms. Methods include radioactively labeled precursor assays, fluorescent chimeric protein tracking, thylakoid translocation studies, subcellular fractionation, affinity purification (TAP-tag), and native PAGE (BN-PAGE) for complex analysis. These approaches enable detailed investigation of chloroplast biogenesis and function, particularly the organellar proteome.
