Het protocol beschrijft de inductie van harige wortels met behulp van Arabidopsis primaire bloeiwijzestengels en Brassica napus hypocotyls. De harige wortels kunnen worden gekweekt en gebruikt als explantaten om transgene planten te regenereren.
Ons onderzoek was gericht op het ontwikkelen van een protocol voor een efficiënte transformatie in Brassicaceae. Met behulp van een transformatie op basis van haarwortels hebben we de transformatie-efficiëntie in Brassica napus drastisch verbeterd, wat cruciaal is voor ons onderzoek. We hebben het protocol ook getest in modelsoorten Arabidopsis thaliana.
We hebben het protocol geoptimaliseerd voor voorjaarscultivars DH12075 in Brassica napus. De volgende uitdaging is het optimaliseren van het protocol van transformatie en regeneratie van wintercultivars in Brassica napus, evenals voor Brassica rapa en Brassica oleracea. Harige wortels kunnen gemakkelijk worden vermeerderd en hun snelle groei maakt een snelle analyse van het betreffende gen kort na de transformatie mogelijk.
Bijgevolg wordt het mogelijk om selectief specifieke harige wortellijnen te regenereren die de gewenste eigenschap vertonen. Bijvoorbeeld een genmutatie die door CRISPR-Cas is geïntroduceerd.
