Mitochondriale overdracht via tunnelende nanobuisjes tussen mesenchymale stamcellen en retinaal pigmentepitheel in vitro

Hier presenteren we een protocol met mitochondriale tracering, directe co-cultuurprocedures van mesenchymale stamcellen (MSC's) en retinale pigmentepitheelcellen (ARPE19), evenals de methoden voor het observeren en statistisch analyseren van de vorming van tunnelende nanobuisjes (TNT) en mitochondriale overdracht om mitochondriale uitwisseling via TNT's tussen MSC's en ARPE19-cellen te karakteriseren.
Onze studie richtte zich op bidirectionele mitochondriale overdracht tussen MSC en de retinale pigmentepitheelcellen. We onderzoeken de overdracht van mitochondriën tussen cellen en bepalen de verhouding van mitochondriale overdracht in twee verschillende richtingen. Het is moeilijk om TNT-gemedieerde mitochondriale overdracht in vivo te detecteren vanwege de dichte seniorenopstelling en uitdagingen om mitochondriën in te voeren en te volgen.
Het is ook moeilijk om de mitochondriën van twee verschillende cellen met verschillende kleuren te labelen. Onze vorige studie observeerde de verschijnselen van TNT-gemedieerde mitochondriale overdracht door MSC mede vast te leggen in retinale cellen, waaronder corneale endotheelcellen, fotoreceptoren en retinale pigmentsignaalcellen in vitro. Functionele mitochondriale overdracht van MSC kan beschadigde en netvliescellen redden.
