The Central Dogma

Główny dogmat biologii molekularnej mówi, że informacja zakodowana w DNA jest przekazywana do RNA, które następnie kieruje syntezą białek w oparciu o te instrukcje.

Po pierwsze, w procesie transkrypcji DNA jest używane jako matryca do syntezy informacyjnego RNA, mRNA, które reprezentuje kopię nici kodującej. Z wyjątkiem tego, że tymidyny są zastępowane uracylami.

Następnie, w procesie translacji u eukariontów, mRNA przemieszcza się do rybosomu. Tutaj kodony, grupy trzech nukleotydów w mRNA, wiążą się z komplementarnymi sekwencjami na transferowych cząsteczkach RNA, tRNA. Każdy z nich jest przyłączony do określonego aminokwasu, w zależności od konkretnego kodonu.

Na przykład kodon CCA wiąże się z tRNA przyłączonym do proliny, podczas gdy AGC wiąże się z tRNA przyłączonym do seryny. W ten sposób kod genetyczny określa kolejność, w jakiej aminokwasy są ułożone w powstałym polipeptydzie. Polipeptydy są często dalej przetwarzane, aby stać się funkcjonalnymi białkami.

Centralny dogmat biologii głosi, że informacja zakodowana w DNA przekazywana jest do informacyjnego RNA (mRNA), który następnie kieruje syntezą białka. Zbiór instrukcji umożliwiających dekodowanie sekwencji nukleotydów mRNA na aminokwasy nazywany jest kodem genetycznym. Uniwersalny charakter tego kodu genetycznego pobudził postęp w badaniach naukowych, rolnictwie i medycynie.

<h4>RNA jest brakującym ogniwem pomiędzy DNA i białkami</h4>

Na początku XX wieku naukowcy odkryli, że DNA przechowuje wszystkie informacje potrzebne do funkcji komórkowych i że białka pełnią większość tych funkcji. Jednak mechanizmy przekształcania informacji genetycznej w białka funkcjonalne pozostawały nieznane przez wiele lat. Początkowo sądzono, że pojedynczy gen jest bezpośrednio przekształcany w kodowane przez niego białko. Dwa kluczowe odkrycia dotyczące komórek eukariotycznych podważają tę teorię: po pierwsze, produkcja białka nie zachodzi w jądrze. Po drugie, DNA nie występuje poza jądrem. Odkrycia te zapoczątkowały poszukiwania cząsteczki pośredniej, która łączy DNA z produkcją białka. Tą cząsteczką pośrednią, znajdującą się zarówno w jądrze, jak i cytoplazmie i związaną z produkcją białka, jest RNA.

Podczas transkrypcji RNA jest syntetyzowane w jądrze, wykorzystując DNA jako matrycę. Nowo zsyntetyzowany RNA ma podobną sekwencję do nici DNA, z tym wyjątkiem, że tymidyna w DNA jest zastąpiona przez uracyl w RNA. U eukariontów ten pierwotny transkrypt jest dalej przetwarzany, usuwając niekodujące regiony białka, zamykając koniec 5’ i dodając ogon poli-A 3’, aby utworzyć mRNA, który jest następnie eksportowany do cytoplazmy.

<h4>Zasady interpretacji sekwencji mRNA stanowią kod genetyczny</h4>

Translacja zachodzi w rybosomach w cytoplazmie, gdzie informacja zakodowana w mRNA jest tłumaczona na łańcuch aminokwasów. Zestaw trzech nukleotydów koduje aminokwas, a te triplety nazywane są kodonami. Zbiór reguł określających, które kodony określają konkretny aminokwas, tworzy kod genetyczny.

<h4>Kod genetyczny jest zbędny</h4>

Białka powstają u eukariontów z 20 aminokwasów. Połączenie czterech nukleotydów w zestawy po trzy daje 64 (43) możliwych kodonów. Oznacza to, że możliwe jest, że pojedynczy aminokwas może być kodowany przez więcej niż jeden kodon. Mówi się, że kod genetyczny jest zbędny lub zdegenerowany. Często, choć nie zawsze, kodony określające te same aminokwasy różnią się jedynie trzecim nukleotydem tripletu. Na przykład wszystkie kodony GUU, GUC, GUA i GUG reprezentują aminokwas walinę. Jednakże AUG jest jedynym kodonem reprezentującym aminokwas metioninę. Kodon AUG jest także kodonem, w którym rozpoczyna się synteza białka i dlatego nazywany jest kodonem start. Redundancja w systemie minimalizuje szkodliwe skutki mutacji. Mutacja (tj. zmiana) w trzeciej pozycji kodonu niekoniecznie musi skutkować zmianą aminokwasu.

<h4>Kod genetyczny jest uniwersalny</h4>

Z kilkoma wyjątkami większość organizmów prokariotycznych i eukariotycznych używa tego samego kodu genetycznego do syntezy białek. Ta uniwersalność kodu genetycznego umożliwiła postęp w badaniach naukowych, rolnictwie i medycynie. Na przykład insulinę ludzką można obecnie wytwarzać na dużą skalę w bakteriach. Odbywa się to przy użyciu technologii rekombinacji DNA. Rekombinowany DNA składa się z materiału genetycznego różnych gatunków. Geny kodujące ludzką insulinę łączą się z bakteryjnym DNA i wstawiają do komórki bakteryjnej. Komórka bakteryjna przeprowadza transkrypcję i translację w celu wytworzenia ludzkiej insuliny zakodowanej w rekombinowanym DNA. Powstałą insulinę ludzką stosuje się w leczeniu cukrzycy.

The central dogma explains how genetic information moves from DNA to RNA to protein. This flow is a key idea in molecular biology. It helps show how cells use genes to make the molecules that carry out work.

DNA stores the instructions for making proteins. During transcription, a gene in DNA is copied into RNA. The RNA then carries that message to the next stage.

In translation, the RNA message is used to build a protein. Proteins do much of the cell’s work, so this process connects genes to traits and cell function. Understanding the central dogma helps students see how information in DNA becomes an active product in the cell.
