Membrane Lipids

W błonie obecne są trzy rodzaje lipidów – fosfoglicerydy, sfingolipidy i sterole.
Fosfoglicerydy, znane również jako glicerofosfolipidy, zawierają szkielet glicerolu z hydrofilową grupą fosforanową i dwoma hydrofobowymi tłuszczowymi łańcuchami acylowymi, zwykle o długości od 16 do 22 atomów węgla.
Niektóre z najczęstszych fosfoglicerydów obecnych w błonie komórkowej to fosfatydylocholina, fosfatydyloetanoloamina, fosfatydyloseryna i fosfatydyloinozytol - utworzony przez przyłączenie choliny, etanoloaminy, seryny i inozytolu do grupy fosforanowej.
Sfingolipidy to lipidy, które zawierają szkielet sfingozyny. Sfingozyna jest aminoalkoholem o długim, nienasyconym łańcuchu węglowodorowym. 
Najprostszym sfingolipidem jest ceramid, który powstaje, gdy cząsteczka kwasu tłuszczowego przyłącza się do grupy aminowej sfingozyny.
Pozostała grupa hydroksylowa ceramidu może tworzyć estry z innymi grupami chemicznymi, takimi jak fosfocholina lub węglowodany, tworząc odpowiednio sfingomielinę lub glikolipidy.
Trzeci rodzaj lipidów błonowych, sterole, takie jak cholesterol, nie mogą samodzielnie tworzyć dwuwarstwy lipidowej. Zamiast tego interkalują w dwuwarstwach fosfolipidowych i działają jako bufor płynności dla błony.

Lipidy są niezbędnym składnikiem wszystkich błon biologicznych. Kwasy tłuszczowe mogą być w pełni nasycone bez podwójnych wiązań, jednonienasycone z tylko jednym podwójnym wiązaniem lub wielonienasycone z wieloma podwójnymi wiązaniami. Glikolipidy obejmują fizjologicznie ważną klasę lipidów błonowych, które są zawsze obecne na zewnętrznej lub zewnątrzkomórkowej stronie błony. U eukariontów znajdują się one zarówno w błonie komórkowej, jak i błonach wewnątrzkomórkowych. Galaktocerebrozyd, prosty glikolipid, występuje obficie w osłonkach mielinowych wraz ze sfingomieliną. Membrane lipids give biological membranes their structure and help set their properties. In mammalian cells, lipids make up about 50% of the membrane on average. The amount can be much lower in the inner mitochondrial membrane or much higher in the myelin sheath around nerve cells.

Several phospholipids are common in mammalian membranes. These include phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, and sphingomyelin. At physiological pH, phosphatidylserine carries a negative charge, while the other three are neutral.

The fatty acids in membrane lipids can differ in carbon length and in saturation. Some are fully saturated, which means they have no double bonds. Others are monounsaturated with one double bond, or polyunsaturated with multiple double bonds.

Glycolipids are an important membrane lipid class that stays on the outer, or extracellular, side of the membrane. In eukaryotes, they are found in both the plasma membrane and intracellular membranes. Galactocerebroside is a simple glycolipid found in large amounts in myelin sheaths along with sphingomyelin.

Gangliosides are the most complex glycolipids. They are negatively charged because they contain sialic acid, and they can make up as much as 10% of the total lipids in nerve cell membranes. Their roles are also studied in viral and genetic diseases. GM1 acts as a cell surface receptor for cholera toxin, and excess GM2 ganglioside is seen in Tay-Sachs disease.

Sterols are another major class of membrane lipids. Plant-specific sterols can make up 30% to 50% of the total lipids in the plant plasma membrane. In eukaryotic membranes, cholesterol is also common and can account for almost 50% of the total lipids. Cholesterol in human cell membranes is slightly different from ergosterol, which is a common lipid in fungal membranes.
