Silne punkty kontaktowe między sąsiednimi komórkami zakotwiczają je ze sobą, tworząc tkanki. Takie połączenia kotwiczące są dwojakiego rodzaju – złącza przylegające i desmosomy. Połączenia przylegające są liczne w tkankach, takich jak nabłonek i śródbłonek, tworząc ciągłą strefę adhezji zwaną pasem adhezyjnym. W innych tkankach, takich jak mięsień sercowy, pojawiają się one w postaci skupisk łączących komórki w celu wywołania skoordynowanego skurczu mięśnia sercowego. 

 Połączenia przylegające są dynamiczne 

 Komórki śródbłonka wyściełające naczynia krwionośne są ściśle ze sobą powiązane, tworząc barierę uniemożliwiającą ucieczkę innych komórek i cząsteczek z krwiobiegu do otaczających tkanek. Jednakże podczas odpowiedzi immunologicznej leukocyty z krwiobiegu muszą przejść przez tę barierę i dotrzeć do tkanki docelowej. Cząsteczki zapalne, takie jak histaminy, i białka sygnalizacyjne, takie jak czynnik wzrostu śródbłonka naczyń (VGEF), wyzwalają dalsze procesy, takie jak fosforylacja i internalizacja kadheryn obecnych na powierzchni komórek śródbłonka. Prowadzi to do rozłączenia połączeń przylegających i zwiększa przepuszczalność śródbłonka, tworząc szczeliny, które umożliwiają przejście komórkom odpornościowym i innym cząsteczkom. Utrzymanie i rozkład połączeń przylegających są zatem dynamicznie regulowane w odpowiedzi na określone bodźce. 

 Połączenia przylegające i infekcja 

 Czynniki zakaźne, takie jak Helicobacter pylori, mogą przedostać się do tkanki gospodarza poprzez rozerwanie połączeń przylegających i przedostanie się przez szczeliny międzykomórkowe. Bakterie te wytwarzają proteazy, które rozcinają zewnątrzkomórkową domenę kadheryn, powodując w ten sposób demontaż połączeń. Kadheryny na powierzchni komórki są również wykorzystywane jako receptory przez bakterie, takie jak Streptococcus pneumoniae, w celu przylegania do nabłonka górnych dróg oddechowych. 

 Jednak najbardziej imponujące porwanie może wykazać Listeria monocytogenes. Bakteria ta wykazuje ekspresję białek ściany komórkowej, które przyłączają E-kadheryny i wyzwalają rekrutację ɑ, β i p120-katenin do tego miejsca. Ta rekrutacja inicjuje dynamiczną polimeryzację aktyny, tworząc występy błonowe, które pochłaniają bakterie w procesie zwanym indukowaną fagocytozą. W ten sposób bakteria przedostaje się do komórki niefagocytarnej i rozmnaża się w żywicielu.

Adherens junctions are strong cell-to-cell anchors that help tissues stay connected. These anchoring junctions work with desmosomes, another type of junction that holds adjacent cells together. In many epithelial and endothelial tissues, adherens junctions form a continuous adhesion belt. In heart muscle, they often appear as clusters that help cells work together for coordinated contraction.

Adherens junctions are also dynamic, especially in the cells that line blood vessels. These endothelial cells form a barrier that normally keeps cells and molecules inside the bloodstream. During an immune response, leukocytes, or white blood cells, must cross this barrier to reach damaged tissue. Inflammatory molecules such as histamines and signaling proteins such as vascular endothelial growth factor, or VEGF, can trigger phosphorylation and internalization of cadherins on the endothelial cell surface. Cadherins are adhesion proteins that help cells stick together. When this happens, adherens junctions break down, permeability increases, and gaps open for immune cells and other molecules to pass through.

Some infections also take advantage of adherens junctions. Helicobacter pylori can disrupt these junctions by producing proteases that cut the extracellular part of cadherins. This breaks the junctions and helps the bacteria move through the spaces between cells. Other bacteria, such as Streptococcus pneumoniae, use cadherins as receptors to attach to the upper respiratory epithelium. Listeria monocytogenes uses a more complex strategy. It binds E-cadherins and recruits alpha-, beta-, and p120-catenins, which starts actin polymerization. This creates membrane protrusions that engulf the bacteria in induced phagocytosis, allowing it to enter a nonphagocytic cell and multiply inside the host.
