Desmosomes

Termin desmosom pochodzi od greckich słów „desmo” i „soma”, co oznacza „ciała adhezyjne”. Strukturę tę po raz pierwszy zaobserwowano pod koniec XIX wieku i opisano ją jako małe, gęste guzki w naskórku. Desmosomy to struktury przypominające guziki, które pomagają tworzyć połączoną sieć włókien pośrednich w komórkach. Połączenia te są niezbędne do utrzymania komórek razem pod wpływem naprężeń mechanicznych i utrzymania integralności tkanki. Desmosomy to kompleksy wielobiałkowe zawierające kadheryny desmosomalne, które są połączone z włóknami pośrednimi za pośrednictwem różnych białek adaptorowych. Kadheryny desmosomalne Kadheryny tworzą zewnątrzkomórkowy region rdzenia desmosomu i wiążą kadheryny na sąsiedniej komórce. W desmosomach występują dwa rodzaje kadheryn – desmogleiny i desmokoliny. Te transbłonowe glikoproteiny posiadają dużą część zewnątrzkomórkową zawierającą cztery powtórzenia kadheryny i jedną zewnątrzkomórkową domenę kotwiczącą, która zakotwicza te białka w błonie komórkowej. Heterodimeryczne pary desmoglein i desmokolin tworzą skupiska w wyniku interakcji cis na tej samej komórce i interakcji trans pomiędzy sąsiednimi komórkami. Ich domeny cytoplazmatyczne mogą różnić się długością między różnymi izoformami ze względu na alternatywny splicing - na przykład u ludzi występują cztery izoformy desmogleiny i trzy izoformy desmokoliny. Białka adaptorowe w desmosomach Domeny cytoplazmatyczne desmosomalnych kadheryn wiążą rodzinę białek pancernika zwanych plakoglobiną i plakofiliną, które tworzą gęstą blaszkę. Białka te mają charakterystyczne powtórzenia ramion, które pełnią funkcję miejsca dokowania dla różnych białek łącznikowych, takich jak desmoplakina. Desmoplakina jest białkiem występującym najczęściej w kompleksie desmosomalnym. Ich koniec karboksylowy zawiera powtórzenia plakiny, które wiążą włókno pośrednie, tworząc w ten sposób połączenie między płytką desmosomalną a cytoszkieletem. Choroby dysfunkcji desmosomu Dysfunkcja desmosomalna zwykle objawia się zaburzeniami serca i skóry – tkanek zazwyczaj poddawanych dużemu obciążeniu mechanicznemu. Na przykład mutacje w genach desmogleiny mogą powodować pogrubienie naskórka i arytmogenną kardiomiopatię prawej komory (ARVC). Ponadto autoprzeciwciała skierowane przeciwko kadherynom desmosomalnym powodują chorobę autoimmunologiczną pęcherzycę zwykłą, w której u pacjentów występują pęcherze naskórkowe z powodu słabej adhezji komórkowej.

Desmosomes are cell junctions that anchor cells together. They are button-like structures that create an interlinked network of intermediate filaments across cells. This strong link helps tissues resist mechanical stress and maintain tissue integrity.

Desmosomes are built from multiple proteins. Their core includes desmosomal cadherins, which are adhesion proteins that bind to cadherins on neighboring cells. The two desmosomal cadherins are desmogleins and desmocollins. These transmembrane glycoproteins have a large extracellular region with four cadherin repeats and one anchor domain that holds them in the cell membrane.

Desmogleins and desmocollins can pair as heterodimers. They form clusters through cis-interactions on the same cell and trans-interactions between adjacent cells. In humans, there are four desmoglein isoforms and three desmocollin isoforms, and their cytoplasmic domains can differ in length because of alternative splicing.

Inside the cell, the cadherins bind armadillo family proteins called plakoglobin and plakophilin. These proteins form a dense plaque. The plaque then recruits desmoplakin, the most abundant protein in a desmosomal complex. Desmoplakin uses its carboxy-terminal plakin repeats to bind intermediate filaments, which connects the desmosome to the cytoskeleton.

Problems with desmosomes often show up in the heart and skin, where cells face high mechanical strain. Mutations in desmoglein genes can cause epidermal thickening and arrhythmogenic right ventricular cardiomyopathy, or ARVC. Autoantibodies against desmosomal cadherins can also cause pemphigus vulgaris, an autoimmune disease that leads to epidermal blisters because cell-cell adhesion becomes weak.
