Kadheryny, klasa cząsteczek adhezyjnych komórek, pośredniczą w selektywnej adhezji między komórkami, a tym samym odgrywają kluczową rolę w tworzeniu i utrzymywaniu struktur tkankowych.
Podczas rozwoju niezróżnicowane komórki wyrażają specyficzne kadheryny na swojej powierzchni, gdy różnicują się w różne typy tkanek, takie jak E-kadheryny na komórkach nabłonka i N-kadheryny na neuronach.
Ponieważ kadheryny na ogół wiążą inne kadheryny tego samego typu, komórki mogą selektywnie przylegać do siebie.
Dodatkowo komórki wyrażające wyższe poziomy kadheryny oddzielają się od tych o niższych poziomach.

Kadheryny stanowią nadrodzinę cząsteczek adhezyjnych do komórek, obejmującą ponad 180 wariantów, przy czym w określonych tkankach zachodzi ekspresja określonej kombinacji typów kadheryn. Kadheryny na ogół wykazują wiązanie homofilne; tj. kadheryny na jednej komórce wiążą się z kadherynami tego samego lub blisko spokrewnionego typu na innej komórce. Zatem komórki tego samego typu mają specyficzne powinowactwo do wiązania się ze sobą i sortowania w skupiska, tworząc tkanki. Sortowanie komórek podczas rozwoju Sortowanie komórek odgrywa zasadniczą rolę podczas embriogenezy i rozwoju, jak wykazały wczesne eksperymenty z zarodkami płazów. Komórki młodych zarodków rozdzielono i pozostawiono do wymieszania. Kiedy tej mieszance pozwolono na agregację, podobne komórki zgrupowały się, tworząc struktury. Struktury te przypominały warstwy embrionalne, takie jak mezoderma, naskórek i cewa nerwowa. Ekspresja kadheryny zmienia się w miarę różnicowania się komórek w trakcie rozwoju, umożliwiając tworzenie złożonych struktur, takich jak cewa nerwowa kręgowców. Jest to charakterystyczna cecha progresji do nowotworu, która umożliwia komórkom nowotworowym ucieczkę i inwazję na inne tkanki. Eksperymentalne blokowanie Twistu w komórkach złośliwego raka piersi zmniejsza ich złośliwość. W przeciwieństwie do tego, normalne komórki nabłonkowe można indukować do poddania się EMT poprzez zwiększenie ekspresji Twist.

Cadherins help cells sort into the right tissues during development. They are a large superfamily of cell adhesion molecules, with more than 180 variants. Different tissues express specific cadherin combinations, which helps cells recognize and stick to the right neighbors.

Cadherins usually bind to the same or a closely related cadherin on another cell. This is called homophilic binding. Because of this matching behavior, cells of the same type tend to group together and form clusters that become tissues.

Cell sorting is important during embryogenesis and development. Early experiments with amphibian embryos showed this clearly. When young embryo cells were separated, mixed, and then allowed to come together again, similar cells clustered and formed structures like the mesoderm, epidermis, and neural tube.

Cadherin expression also changes as cells differentiate. During vertebrate neural tube formation, neural plate cells switch from E-cadherin to N-cadherin as the neural groove forms from the ectoderm. Cells at the neural plate border also begin expressing other cadherins, such as cadherin 6B. These changes help bring the two sides together and support pinching off of the neural tube.

Cadherin-mediated adhesion also helps maintain tissue homeostasis in multicellular organisms. In cancer, this adhesion can break down. Many epithelial tumor cells lose some or all E-cadherin expression, often with help from transcription regulators. One example is Twist, which blocks E-cadherin expression and reduces cell adhesion to neighboring cells.
