Chromosome Duplication

Materiał genetyczny, DNA, w komórce eukariotycznej istnieje wewnątrz jądra jako liniowe struktury zwane chromosomami.
W chromosomach dwuniciowy DNA owija się głównie wokół oktamerycznych białek jądrowych zwanych histonami, tworząc nukleosomy. Kilka białek niehistonowych również wiąże się z DNA, pomagając w pakowaniu DNA.
Te nukleosomy i białka niehistonowe układają się, tworząc cewki, które dalej składają się w pętle w układzie, tworząc upakowaną chromatynę. Stopień zagęszczenia chromatyny umożliwia zmieszczenie się bardzo długiego DNA w jądrze. Podczas fazy M chromatyna dalej się zwija, tworząc skondensowane chromosomy.
Duplikacja chromosomów polega na duplikacji całej struktury chromatyny, więc DNA i białka związane z chromatyną są zduplikowane. Produkcja histonów jest zwiększana w celu wytworzenia dodatkowych białek wymaganych do upakowania nowo zsyntetyzowanego DNA w chromosomy.
Replikacja dokładnej struktury chromatyny zduplikowanego chromosomu ma kluczowe znaczenie dla regulacji genów. 
W obrębie chromosomów poziom upakowania chromatyny nie zawsze jest jednolity. Chromosomy zawierają regiony z ciasno upakowaną chromatyną zwaną heterochromatyną i regiony z luźno upakowaną chromatyną zwaną euchromatyną.
DNA heterochromatyny jest niedostępne dla maszynerii transkrypcyjnej, a zatem geny w tych regionach nie są regularnie transkrybowane. W przeciwieństwie do tego, DNA w euchromatynie jest bardziej dostępne, a zatem geny w tych regionach mogą być transkrybowane. 
To strukturalne i funkcjonalne znaczenie chromatyny sprawia, że kluczowe jest, aby cała struktura chromatyny była dokładnie odtwarzana podczas duplikacji chromosomów.

Proces duplikacji chromosomów podczas podziału komórki wymaga rozerwania całego genomu i ponownego złożenia chromatyny. Struktura chromatyny musi być dokładnie skopiowana ponownie składana i utrzymywana w komórkach potomnych, aby zapewnić propagację linii. Podstawową jednostką chromatyny jest nukleosom, składający się z DNA owiniętego wokół oktamerycznych białek histonowych i krótkich odcinków DNA łącznikowego oddzielającego poszczególne nukleosomy. Białka histonowe w nukleosomie mają swoje N-końce wystające z rdzenia, zapewniając miejsca dla różnych modyfikacji kowalencyjnych, które regulują strukturę i funkcję chromatyny. Podczas replikacji, gdy DNA się rozwija, nukleosomy rodzicielskie ulegają rozerwaniu i uwalniane są białka histonowe. W miarę postępu replikacji i tworzenia nici potomnych histony macierzyste i dodatkowe białka histonowe syntetyzowane podczas fazy S łączą się, umożliwiając utworzenie nukleosomów. Modyfikacje potranslacyjne ogona histonowego służą do utrzymania aktywnego lub nieaktywnego stanu transkrypcji. Specyficzne modyfikacje ogona histonowego mogą złagodzić poziom zagęszczenia DNA, ułatwiając destabilizację i przemieszczanie nukleosomów, zapewniając dostęp maszynerii transkrypcyjnej w celu wpływania na ekspresję genów.
