Forces Acting on Chromosomes

Podczas mitozy dwukierunkowe chromosomy oscylują, aby ustabilizować swoje przyczepy wrzeciona, a następnie gromadzą się wzdłuż płytki metafazowej na równiku wrzeciona mitotycznego.
Na chromosomy działają wiele sił po ich przyłączeniu do wrzeciona mitotycznego.
Duża siła skierowana do bieguna wywierana wzdłuż mikrotubuli kinetochoru, w wyniku depolimeryzacji końca plus, przyciąga kinetochor i związany z nim chromosom w kierunku bieguna wrzeciona. W kinetochorze kompleksy białkowe Ndc80 łączą kinetochor z mikrotubulą poprzez wiele przyłączeń o niskim powinowactwie wzdłuż boków mikrotubul.
Podczas depolimeryzacji mikrotubul z plusem końcówki Ndc80 pękają i przekształcają się w nowych miejscach, aby utrzymać połączenie kinetochor-mikrotubula. Mechanizm stopniowo przyciąga chromosom w kierunku bieguna wrzeciona, w miarę jak mikrotubula skraca swoją długość.
Druga siła w kierunku bieguna wynika ze strumienia mikrotubul. Depolimeryzacja mikrotubuli z minusem końca generuje strumień skierowany na minus koniec, powodując ruch mikrotubuli w kierunku bieguna wrzeciona. Koordynacja polimeryzacji końca dodatniego kompensuje depolimeryzację końca ujemnego, umożliwiając mikrotubulom zachowanie swojej długości.

Podczas mitozy ruchy chromosomów zachodzą w wyniku wzajemnego oddziaływania wielu sił na poziomie piconewtona. W prometafazie siły te pomagają w składaniu lub gromadzeniu chromosomów w płaszczyźnie równikowej, ostatecznie prowadząc do ich wyrównania na płytce metafazowej. Siły działające na chromosomy są zależne od przestrzeni i czasu; dlatego różnią się w zależności od położenia chromosomów w miarę przechodzenia komórki przez mitozę. Mikrotubule i białka motoryczne wywierają na chromosomy dwa rodzaje sił – biegunowe i antybiegunowe, znane również jako siły wyrzutu polarnego. Depolimeryzacja mikrotubul kinetochorowych generuje siłę biegunową i przyciąga chromosom w stronę bieguna wrzeciona. Natomiast polimeryzacja mikrotubuli kinetochorowej prowadzi do sił wyrzutu polarnego, które popychają chromosom w kierunku równika komórki. Białka motoryczne skierowane na koniec plus mikrotubul, takie jak chromokinezy i kinezyna-7, również wytwarzają siły wyrzutu polarnego, wpychając chromosomy w kierunku równika komórki. Jednoczesne, ale nierówne działanie sił biegunowych i wyrzutowych powoduje oscylację chromosomów podczas prometafazy; jednakże podczas metafazy na dwuorientowane chromatydy siostrzane działają równe, ale przeciwne siły. Wytwarza to wystarczające napięcie, aby wyciszyć ścieżkę punktu kontrolnego zespołu wrzeciona i umożliwia komórkom przejście do anafazy. W anafazie siły biegunowe działają na chromatydy siostrzane, powodując ich skuteczną segregację do komórek potomnych.
