Pod koniec metafazy dwukierunkowe chromosomy wyrównują się na płytce metafazowej. Podczas tej fazy kompleksy pierścieniowe białek kohezyny utrzymują chromatydy siostrzane razem w regionie centromerów i zapobiegają ich rozerwaniu.
Progresja metafazy do anafazy jest wyzwalana przez indukowaną cykliną Cdk fosforylację wielopodjednostkowego, enzymu ligazy ubikwityny – kompleksu promującego anafazę, znanego również jako cyklosom lub APC / C. Fosforylowany APC / C wiąże się z białkiem Cdc20, tworząc aktywny kompleks.
Aktywny kompleks APC/C rozpoznaje białko hamujące zwane sekuryną, które jest związane z enzymem proteazy zwanym separazą. Przed tym rozpoznaniem sekuryna hamuje aktywność separazy.
Aktywny kompleks APC/C znakuje sekurynę białkiem ubikwityną, kierując ją do degradacji proteasomalnej. Zniszczenie sekuryny uwalnia separazę.

Inaktywacja Cdks umożliwia enzymom fosfatazy defosforylację separazy i zapobiega ponownej fosforylacji separazy za pośrednictwem Cdk. Defosforylacja pozwala enzymowi separazy na rozszczepienie kompleksu pierścieniowego kohezyny, który utrzymuje razem chromatydy siostrzane.
Rozszczepienie kohezyny powoduje bezwzględną utratę kohezji chromatydy siostrzanej, co oznacza przejście z metafazy do anafazy. Utrata kohezji pozwala chromatydom siostrzanym na rozdzielenie się i przemieszczenie do przeciwległych biegunów wrzeciona.

Jednakże resztkowe połączenia kohezynowe pozostają, aby utrzymać razem chromatydy siostrzane aż do przejścia z metafazy do anafazy. Resztkowe połączenie zapobiega przedwczesnemu rozdzieleniu chromatyd siostrzanych, blokując ryzyko aneuploidii w komórkach potomnych. Na początku anafazy aktywowana jest separaza, enzym proteolityczny. Aktywowana separaza rozszczepia podjednostkę Scc1 pozostałych pierścieni kohezyny, powodując całkowitą utratę spójności. Tylko wtedy, gdy każdy chromosom jest prawidłowo ustawiony na wrzecionie mitotycznym, SAC zostaje dezaktywowany, aby umożliwić fosforylację i aktywację podjednostek regulatorowych APC/C - CDC20 i CDH1. Po drugie, CDC20 i CDH1 są regulowane przez różne mechanizmy. Gdy APC/C-CDC20 jest aktywny, CDH1 ulega hamującej fosforylacji przez cyklinę B/Cdk1, uniemożliwiając mu wiązanie się z APC/C.
