Barwienie w mikroskopii zwiększa widoczność mikroorganizmów poprzez rozróżnienie ich struktur wewnętrznych i zewnętrznych. Proste barwienie wykorzystuje pojedynczy barwnik do podkreślenia rozmiaru, kształtu i podstawowych cech komórki. Utrwalanie jest kluczowym krokiem, który zachowuje morfologię komórek i unieruchamia struktury przed barwieniem. Utrwalanie ciepła jest powszechnie stosowane w próbkach bakteryjnych i archeonów w mikroskopii świetlnej. Aby wykonać proste barwienie, cienki rozmaz kultury drobnoustrojów rozprowadza się na szkiełku, suszy na powietrzu i utrwala na gorąco. Szkiełko jest następnie barwione podstawowym barwnikiem - takim jak fiolet krystaliczny, błękit metylenowy lub safranina - przez 1 do 2 minut. Po zabarwieniu szkiełko spłukuje się wodą w celu usunięcia nadmiaru barwnika i osusza papierem chłonnym. Barwniki zasadowe z dodatnio naładowanymi chromoforami wiążą się z ujemnie naładowanymi składnikami komórek, takimi jak kwasy nukleinowe i ściany komórkowe, poprzez przyciąganie elektrostatyczne. W ten sposób powstaje jaskrawo zabarwiona komórka na wyraźnym tle, dzięki czemu morfologia i układ drobnoustrojów są łatwiejsze do zaobserwowania.

Simple Staining Technique

Simple staining helps reveal the size, shape, and arrangement of microorganisms under the microscope. It uses one dye to color the whole cell. That creates strong contrast between the cells and the background.

A careful preparation step comes first. A small drop of microbial culture is spread on a clean glass slide to make a thin smear. The smear is air-dried and then heat-fixed, which means it is briefly passed over a flame.

Heat fixation keeps the cells attached to the slide. It also helps prevent autolysis, or self-destruction of the cells, and preserves their shape. In addition, it can improve dye uptake by changing the permeability of the cell wall.

After fixation, the slide is covered with a basic dye such as crystal violet, methylene blue, or safranin for 1 to 2 minutes. These dyes have positively charged chromophores that bind to negatively charged parts of the cell, including nucleic acids and the cell wall. The slide is then rinsed with water and blotted dry with absorbent paper.

The result is brightly stained cells on a clear background. This makes it easier to observe microbial morphology and arrangement. Simple staining does not show detailed structures such as organelles, and it does not tell Gram-positive and Gram-negative bacteria apart. Even so, it is a useful first step in microbiology and often comes before more advanced staining methods.
