A lethal allele is a mutation in an essential gene that results in the death of an organism. Essential genes are necessary for survival, and lethal alleles disrupt their normal function. When inherited, lethal alleles prevent organisms from developing properly, often causing death during embryonic development or early life stages.

Dominant lethal alleles cause death in both homozygotes and heterozygotes carrying the mutation, while recessive lethal alleles only kill homozygotes. Dominant lethal alleles are rarely observed in populations because affected individuals die quickly. Recessive lethal alleles persist longer because heterozygotes survive and can reproduce, passing the allele to offspring.

Cuénot observed a 2:1 ratio of yellow to gray mice because homozygous yellow embryos died during development. The yellow allele is lethal when homozygous, eliminating one-quarter of expected offspring. This left only heterozygous yellow mice and homozygous gray mice surviving, producing the unexpected 2:1 phenotypic ratio instead of the typical Mendelian 3:1 ratio.

The agouti gene determines coat color in mice by controlling melanin distribution through agouti-signaling protein. The wild-type allele produces gray-brown coat color, while the mutant allele produces yellow coat color. The mutant allele is also associated with yellow mouse obesity syndrome, characterized by early obesity and tumor development.

Castle and Little discovered that one-quarter of offspring from heterozygous yellow crosses died during embryonic development. These failed embryos were homozygous for the mutant agouti allele, confirming it as a recessive lethal. This explained why Cuénot never observed homozygous yellow mice and why the breeding results deviated from expected Mendelian ratios.

Huntington's disease is a dominant lethal allele causing a fatal neurological disorder. Unlike most dominant lethal alleles, disease onset occurs around age 40, allowing heterozygotes to survive reproductive years and pass the mutation to offspring. This delayed lethality enables the allele to persist in populations, unlike lethal alleles causing early death.

Achondroplasia causes short-limbed dwarfism when a single mutant allele is present, making it dominant for the disease phenotype. However, homozygous individuals carrying two mutant alleles die during embryonic development, making the lethality recessive. This demonstrates that dominance for disease expression differs from dominance for lethality.
