Lipid-proximal Interactions

Lipid-proximal interactions are molecular contacts between proteins, peptides, or other biomolecules and nearby lipids, linking membrane composition to biochemical function. They arise when electrostatic forces, hydrophobic contacts, or specific lipid-binding sites position a molecule at the membrane surface or within the bilayer, potentially altering its conformation and activity. In biochemistry, analyzing these interactions helps explain membrane recruitment, protein organization, enzymatic regulation, and cell signaling. Experimental and computational approaches can identify lipid-binding regions and assess how changes in lipid composition affect molecular behavior, providing insight into membrane-associated pathways and supporting research on disease mechanisms and therapeutic targets.

This paper describes a method for assessing the interactions and assemblies of integral membrane proteins in vitro with various partner factors in a lipid-proximal environment.

Microscale Thermophoresis to Study Protein-Lipid Interactions in Solution

2025

This video demonstrates microscale thermophoresis for studying the protein-lipid interaction. The assay detects the interaction between molecules by quantifying the thermophoretic movement of fluorescent-labeled proteins in response to a temperature gradient. The fluorescent molecule is mixed with different concentrations of the non-fluorescent lipid molecules, and the mixture of molecules in the solution is loaded into capillaries. A temperature gradient is applied to the samples in the...

Amplified Luminescent Proximity Homogeneous Assay: A Bead-Based Proximity Assay to Screen Small Molecules Inhibiting Protein-Protein Interactions

In this video, we demonstrate the amplified luminescent proximity homogeneous assay—a bead-based proximity assay. It utilizes homogeneously-sized acceptor and donor beads, immobilized with two proteins that tend to interact to screen potential small molecules inhibiting protein interactions.

Detection of Protein-Protein Interactions in Drosophila Larvae Using a Proximity Ligation Assay

2025

This video demonstrates the visualization of protein-protein interactions at the Drosophila neuromuscular junction (NMJ) using Proximity Ligation Assay (PLA). After isolating the body walls, interacting proteins are probed with primary antibodies and PLA-conjugated secondary antibodies. The proximity of the interacting proteins facilitates the ligation of probes with connector oligonucleotides, forming circular DNA, which is then amplified by rolling circle amplification. Fluorescent probes...

Proximity Ligation Assay to Study In Situ Protein-Protein Interactions

2025

This video describes the proximity ligation assay to study in situ protein-protein interactions in nematode cells. The close proximity of the proteins of interest generates the circular DNA template with the help of a PLA probe attached to a secondary antibody. The probe is further amplified using the amplification solution, which produces a long single-stranded DNA followed by the hybridization of the detector probe, enabling the localization of protein-protein interactions in cells.
