Nitride Membrane

Nitride membranes are thin, nitrogen-containing ceramic films that act as mechanically stable barriers or supports in microscale and nanoscale devices. Their function arises from a combination of low thickness, controlled pore structure, and tunable surface chemistry, which can regulate molecular transport, separate fluids, or support electrical and optical sensing while resisting chemical and mechanical stress. In bioengineering, silicon nitride membranes are used in microfluidic platforms, nanopore biosensors, and interfaces for studying cells or biomolecules, where membrane geometry and surface modification influence permeability, analyte capture, and signal quality. These properties support compact diagnostic devices and other tools for biological measurement.

Presented here is a protocol for cell culture on silicon nitride membranes and plunge-freezing prior to X-ray fluorescence imaging with a synchrotron cryogenic X-ray nanoprobe. When only room temperature nano-analysis is provided, the frozen samples can be further freeze-dried. These are critical steps to obtain information on the intracellular elemental composition.
