Split Luciferase

Split luciferase is a bioluminescence-based technique that detects molecular proximity, most commonly protein-protein interactions, by dividing a luciferase enzyme into two inactive fragments. Researchers fuse each fragment to a protein of interest; when the proteins interact or approach one another, the luciferase domains complement and reconstitute an active enzyme that emits light after reacting with its substrate. Because the signal can be measured quantitatively, split luciferase supports real-time analysis of cellular interactions, signaling pathways, protein dynamics, and changes caused by drugs or other experimental conditions. Its sensitivity and compatibility with live-cell assays make it valuable for biological research and screening.

This video demonstrates the split luciferase complementation assay to detect protein-protein interactions. In this assay, the proteins of interest are tagged to small and large fragments of the luciferase enzyme. When the proteins interact, the large and small fragments combine to form an active enzyme complex, which in the presence of a specific substrate, releases bright luminescence that can be measured.

Here, we present a method for screening anti-hepatitis B viral agents that inhibit the HBx-DDB1 interaction using a split luciferase assay system. This system allows easy detection of protein-protein interactions and is suitable for identifying inhibitors of such interactions.

We have established a split-luciferase reassembly assay to monitor the endoplasmic reticulum-mitochondria contacts in live cells. Using this assay, we describe a protocol to quantitatively measure the level of these inter-organelle couplings in HEK293T cells, under the condition of chemical treatment.

A Split-Luciferase Complementation Assay for Temporally Resolved Measurement of Tau Clearance in Microglia

We describe a sensitive, time-resolved quantitative platform to measure clearance of extracellular tau in human induced pluripotent stem cell (iPSC)-derived microglia using a microplate-based luminescence system. This approach enables sensitive detection of tau degradation kinetics in a microwell format, allowing for high-throughput interrogation of tau handling under various experimental conditions.
