Elongation Factors

Elongation factors are proteins that drive and regulate the elongation stage of protein synthesis, making them essential for translating genetic information into functional proteins. During translation, GTPase elongation factors use GTP hydrolysis to deliver aminoacyl-tRNAs to the ribosome, promote correct codon-anticodon pairing, and support movement of the ribosome along messenger RNA; related factors release spent components and reset the cycle. In biology, studying these factors clarifies how cells control translation speed, accuracy, and energy use, while also informing research on antibiotics, genetic disease, and cellular responses to stress.

Here we detail protocols specifically designed to monitor morphogenic defects that occur during early and late phases of embryonic elongation of the nematode Caenorhabditis elegans. Synthesis of custom plasmids is labor and time consuming. This protocol describes the use of Gibson assembly cloning to reduce the work and duration of custom DNA cloning procedure.
