Tissue Chopper Method

The Tissue Chopper Method is a mechanical technique for cutting fresh tissue into thin, relatively uniform sections or explants while retaining aspects of its native three-dimensional organization. A movable blade repeatedly advances through tissue at controlled intervals, producing pieces that can be transferred to culture and maintained under defined experimental conditions. In cancer research, these tissue slices support organotypic models that preserve interactions among tumor cells, stromal cells, and extracellular matrix more effectively than isolated cell cultures. Researchers use the method to examine tumor biology, treatment responses, and microenvironmental effects, helping connect laboratory findings with tissue-level disease behavior.

Here we present a hydrophobic tissue clearing method that allows for the viewing of target molecules as part of intact brain structures. This technique has now been validated for F344/N control and HIV-1 transgenic rats of both sexes.

Here we present, and contrast two protocols used to decellularize plant tissues: a detergent-based approach and a detergent-free approach. Both methods leave behind the extracellular matrix of the plant tissues used, which can then be utilized as scaffolds for tissue engineering applications.
