Hydrophobic Colloids

Hydrophobic colloids are dispersions in which particles of a water-insoluble substance remain suspended in a liquid despite having little affinity for the dispersion medium. They form when finely divided particles disperse through a continuous phase and are stabilized mainly by surface charge, whose electrostatic repulsion limits aggregation; electrolytes can screen these charges and cause coagulation. In chemistry, hydrophobic colloids help explain sol stability, adsorption, and precipitation behavior, with examples including metal sols and sulfur sols. Understanding their formation and destabilization supports water treatment, formulation, and separation processes.

In this video, we demonstrate the purification of calcium-binding protein from a dialyzed cell lysate through calcium-dependent hydrophobic interaction chromatography. The calcium-binding proteins expose a hydrophobic region upon binding with calcium, facilitating interaction with a hydrophobic group on resin. Later these proteins are eluted using calcium chelator EDTA that reverses the interaction.
