Menadione

Menadione is a synthetic naphthoquinone, formally 2-methyl-1,4-naphthoquinone, and is commonly known as vitamin K3. Its conjugated quinone structure enables reversible oxidation and reduction: menadione accepts electrons to form a semiquinone or hydroquinone intermediate, and redox cycling can generate reactive oxygen species in the presence of oxygen and suitable reducing agents. These chemical properties make menadione useful in studying quinone reactivity, electron-transfer processes, oxidative stress, and vitamin K-related biochemistry. In chemistry and biomedical research, it also serves as a model compound for examining redox-active molecules and their effects on cellular and microbial systems.

An In Vitro Assay to Evaluate the Effect of Antibodies on Candida tropicalis Biofilm

2025

This video demonstrates an in vitro technique to study the effect of antibodies on fungal biofilms. The pathogenic fungus Candida tropicalis secretes a virulence factor secreted aspartyl proteinase 2 (Sap2), facilitating biofilm formation. Upon introducing a heat-inactivated serum containing Sap2-specific antibodies that inhibit biofilm maturation, the viability of the fungal cells is assessed using a chromogenic assay.

An Assay to Study the Impact of Photodynamic Treatment with a Photosensitizer on Macrophage Metabolic Activity

Sebolai11Department of Microbiology and Biochemistry, University of the Free State. The video demonstrates a technique for evaluating the influence of photodynamic treatment (PDT) with a photosensitizer on the metabolic activity of murine macrophages. The cells undergo treatment with the photosensitizer followed by exposure to UV light, initiating PDT. This process triggers the photosensitizer to generate reactive oxygen species...

Fluorescence-Based Viability Assessment of Oral Anaerobic Bacteria

Source: Calatayud Arroyo, M., et al. Assessing the Viability of a Synthetic Bacterial Consortium on the In Vitro Gut Host-microbe Interface. J. Vis. Exp. (2018)The video demonstrates a method to evaluate the viability of anaerobic oral bacteria using fluorescent dyes and flow cytometry. It begins with culturing bacteria in oxygen-free broth, followed by resuspending a single colony from a blood agar plate. After incubation and dilution, the culture is stained with membrane-permeable and...
