Nitridation Process

Nitridation is a chemical process that introduces nitrogen into a material or compound, often to alter its composition, structure, and properties. During nitridation, reactive nitrogen species combine with the target substance or diffuse into its surface under controlled conditions of temperature, pressure, and gas composition, forming metal nitrides or nitrogen-containing compounds. In chemistry and materials science, the process supports surface hardening of steels, synthesis of ceramic materials, modification of catalysts, and preparation of semiconductor-related compounds. Its outcomes depend on the nitrogen source, reaction environment, and substrate, making process control essential for achieving uniform composition and desired performance.

Presented here is a protocol for cell culture on silicon nitride membranes and plunge-freezing prior to X-ray fluorescence imaging with a synchrotron cryogenic X-ray nanoprobe. When only room temperature nano-analysis is provided, the frozen samples can be further freeze-dried. These are critical steps to obtain information on the intracellular elemental composition.
