Proton Carbon Correlation

Proton-carbon correlation is a two-dimensional nuclear magnetic resonance (NMR) method that reveals relationships between hydrogen and carbon atoms, helping identify molecular structure and connectivity. During the experiment, magnetization transfers between coupled ¹H and ¹³C nuclei through scalar, or J, coupling, producing cross-peaks that connect their chemical shifts; one-bond correlations commonly arise in HSQC, while longer-range correlations can be observed with HMBC. These spectra help assign signals, distinguish similar environments, and map carbon frameworks in organic compounds. Proton-carbon correlation supports structure elucidation, reaction monitoring, metabolite identification, and the characterization of pharmaceuticals, natural products, and other chemically complex samples.
