Real-time Binding

Real-time binding is the continuous measurement of interactions between chemical or biological molecules as they associate and dissociate, rather than assessing binding only at the experiment’s endpoint. In chemistry, an instrument tracks a signal that changes when one binding partner interacts with another, allowing researchers to determine association and dissociation rates, binding strength, and equilibrium behavior under defined conditions. These measurements help characterize complexes such as protein–ligand, receptor–drug, and antibody–antigen pairs. By revealing binding kinetics and selectivity, real-time binding analysis supports mechanism studies, molecular screening, assay development, and the optimization of therapeutic or diagnostic compounds.

This protocol presents a method for measuring adenine nucleotide binding to receptors in real time in a cellular environment. Binding is measured as Förster resonance energy transfer (FRET) between trinitrophenyl nucleotide derivatives and protein labeled with a non-canonical, fluorescent amino acid.

Real Time RT-PCR

2021

Real-time reverse transcription-polymerase chain reaction, or Real-time RT-PCR, is an analytical tool used to determine the expression level of target genes. The method involves converting mRNA to complementary DNA with the help of an enzyme known as reverse transcriptase, followed by the PCR amplification of the cDNA. These two processes can be performed simultaneously in a single tube or separately as a two-step reaction. The real-time quantification of the number of amplified products is...
