Stacking Technique

The stacking technique is an analytical method that concentrates sample components into a narrow zone before separation, improving the resolution and detectability of chemical analytes. In electrophoretic applications, it uses differences in conductivity and electric-field strength between the sample, leading electrolyte, and terminating electrolyte to compress migrating ions into a focused band. This process can increase analyte concentration without changing the sample’s chemical identity, making low-abundance compounds easier to detect. Stacking is therefore valuable in capillary electrophoresis and related separation methods, where it supports sharper peaks, improved sensitivity, and more reliable qualitative or quantitative analysis.

Scaling of Engineered Vascular Grafts Using 3D Printed Guides and the Ring Stacking Method

Scalable engineered blood vessels would improve clinical applicability. Using easily sizable 3D-printed guides, rings of vascular smooth muscle were created and stacked into a tubular form, forming a vascular graft. Grafts can be sized to meet the range of human coronary artery dimensions by simply changing the 3D-printed guide size.

Here we provide a detailed procedure for large-scale production of research-grade AAV vectors using adherent HEK 293 cells grown in cell stacks and affinity chromatography purification. This protocol consistently yields >1 x 1013 vector genomes/mL, providing vector quantities appropriate for large animal studies.

The key steps of living anionic polymerization of phenyl glycidyl ether (PheGE) on methoxy-polyethylene glycol (mPEG-b-PPheGE) are described. The resulting block copolymer micelles (BCMs) were loaded with doxorubicin 14% (wt%) and sustained release of drug over 4 days under physiologically relevant conditions was obtained.
