Neutrophil Purification

Neutrophil purification is the laboratory process of isolating neutrophils from whole blood or other cell mixtures for controlled study of innate immune responses. It commonly combines density-gradient centrifugation with red blood cell removal or negative immunomagnetic selection, separating neutrophils according to density or selectively depleting unwanted leukocytes while preserving cell viability and function. Purified neutrophils support research on phagocytosis, chemotaxis, degranulation, oxidative burst, and pathogen killing in immunology and infection studies. Reliable isolation enables researchers to compare inflammatory responses, investigate host-pathogen interactions, and evaluate how drugs or microbial factors alter neutrophil behavior.

Rapid Magnetic-microbead Method for Efficient Purification of Low-density Neutrophils

Low-density neutrophils (LDN) increase significantly in several diseases. LDN are usually isolated through cell sorting. We present a practical method to obtain pure and viable LDN. After density-gradient centrifugation of peripheral blood, cells are incubated with magnetic microbeads, and then LDN are separated through magnetic columns.

A high yield method for one-step negative purification of recombinant Helicobacter pylori neutrophil-activating protein (HP-NAP) overexpressed in Escherichia coli by using diethylaminoethyl resins in batch mode is described. HP-NAP purified by this method is beneficial for the development of vaccines, drugs, or diagnostics for H. pylori-associated diseases.

We describe a protocol for isolation and purification of neutrophils from mouse bone marrow by density gradient centrifugation and for neutrophil labeling using CellTracker dyes. This represents a simple, fast, reproducible and economical method for obtaining large numbers of neutrophils for downstream functional studies or adoptive transfer and tracking experiments.
