Photometer

A photometer is an instrument that measures the intensity of light to quantify properties of a sample, making it useful for detecting and comparing biological substances. It works by directing light through or onto a sample and measuring transmitted, absorbed, or emitted light; in absorbance assays, the signal changes with analyte concentration according to the Beer-Lambert relationship under defined conditions. In immunology and infection research, photometers support colorimetric ELISAs, protein assays, and measurements of microbial growth or metabolic activity. These readings help researchers estimate antibody or antigen levels, monitor pathogen-associated changes, and standardize quantitative experiments.

2019

The goal of the methods presented here is to measure aerosol optical thickness of the atmosphere. The sun photometer is pointed at the sun and the largest voltage reading obtained on an in-built digital voltmeter is recorded. Atmospheric measurements such as barometric pressure and relative humidity are also performed.

Minimum Erythematous Dose Assay: A Method for Measuring UV Sensitivity in Mouse Models

2023

This video describes a method to measure UV sensitivity in a humanized mouse model—a mouse model that mimics human skin. We determine the minimal erythematous dose (MED) of UV radiation that can cause erythema or edema on the skin. The method can help to find out the efficacy of drugs used to reduce UV sensitivity.

Mass Photometry to Study Antigen-Antibody Interactions on a Single-Molecule Level

2025

In this video, we demonstrate the use of mass photometry to study antigen-antibody interactions in a flow chamber. The focused light scatters differently based on the mass of the molecules. A detector collects the scattered light and generates the interference pattern as spots of varied intensities, which correlate with the molecular masses.

Bacterial Cell Staining and Preparation for Immunological Studies

2025

This video demonstrates the staining and preparation of bacterial cells for immunological studies using red fluorescent dye. The stained bacteria are diluted and resuspended in a growth medium to achieve the desired bacterial concentration for host cell infection, representing the multiplicity of infection.

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.
