Surgical Model Preparation

Surgical model preparation is the standardized planning and execution of operative procedures used to create reliable in vivo models for studying physiology, disease, immunity, and infection. It typically combines model selection, anesthesia, aseptic handling, surgical access, and postoperative monitoring so that tissue injury, microbial exposure, and host responses can be controlled and measured consistently. In immunology and infection research, these steps establish reproducible conditions for examining pathogen dissemination, inflammation, immune-cell recruitment, and treatment effects while protecting animal welfare and reducing experimental variability. Careful preparation strengthens data quality and supports comparisons across experiments, from localized infection models to studies of systemic immune responses.

This article demonstrates surgical procedures of gastroesophageal reflux with mice. These models are useful tools for research on mechanisms and treatment of gastroesophageal reflux disease and potentially Barrett’s esophagus and esophageal adenocarcinoma.

MSC Seeded Scaffold Preparation: A Technique to Prepare Scaffold Seeded with Mesenchymal Stem Cells for Surgical Implantation into Murine Model

2025

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.

All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Preoperative Preparation The initial assessment should include observation of alertness, posture, gait, and grooming behavior, as changes in these parameters may indicate underlying health issues. Body weight should be recorded...
