Dormant Colony Quantification

Dormant colony quantification is a microbiological method for measuring viable cells that remain metabolically inactive or grow too slowly to form colonies under standard culture conditions, a state relevant to persistent infection and treatment failure. The approach typically exposes samples to defined conditions that preserve or induce dormancy, then uses resuscitation or optimized culture conditions to enable colony formation and counts the resulting colony-forming units as an estimate of the dormant population. In medicine, this measurement helps characterize microbial persistence, evaluate antimicrobial activity against nonreplicating cells, and assess strategies for detecting or eliminating infection reservoirs that conventional culture may underestimate.

ScanLag: High-throughput Quantification of Colony Growth and Lag Time

ScanLag is a high-throughput method for measuring the delay in growth, namely lag time, as well as the growth rate of colonies for thousands of cells in parallel. Screening using ScanLag enables the discrimination between long lag-time and slow growth at the level of a single variant.
