Compound Bioactivity Screening

Compound bioactivity screening is a systematic method for determining whether chemical compounds produce measurable effects in biological systems, helping researchers identify promising molecules and characterize their activity. In a typical assay, compounds are applied across concentrations to purified neural targets, cultured neurons, or other model systems, while responses such as receptor signaling, ion-channel activity, cell viability, or synaptic function are quantified against appropriate controls. These measurements help estimate potency, selectivity, and toxicity before active compounds advance to secondary assays. In neuroscience, screening supports the discovery of modulators for neurotransmitter receptors and disease-relevant pathways, linking chemical activity to neuronal mechanisms and potential therapeutic development.

Toll-like receptor (TLR) signaling plays an important role in the pathophysiology of many human inflammatory diseases, and regulating TLR responses by bioactive nanoparticles is anticipated to be beneficial in many inflammatory conditions. THP-1 cell-based reporter cells provide a versatile and robust screening platform for identifying novel inhibitors of TLR signaling.

A Fluorescence-Based Assay Using Engineered Lymphocytes for Screening Immunomodulatory Compounds

2025

This video demonstrates the screening of immunomodulatory compounds via fluorescent-based assay. Transgenic mouse-derived T lymphocytes with green fluorescent protein expression cassettes are treated with these compounds. The enhanced and reduced GFP signals enable the identification of the compounds' stimulatory and inhibitory effects, respectively.

This video demonstrates a viral attachment assay for screening antiviral compounds. Using genetically modified hepatitis C virus and luciferase, it assesses inhibitory effects on viral attachment, as indicated by lower light intensity in co-treated wells compared to wells treated with the virus alone.
