Controlled Amyloid Beta Release

Controlled amyloid beta release is the experimental regulation of when and how much amyloid beta (Aβ) is produced and secreted by neurons or model systems, enabling precise study of this Alzheimer’s disease-related peptide. Aβ forms when amyloid precursor protein undergoes sequential cleavage by beta-secretase and gamma-secretase, after which the peptide can move through secretory or endosomal pathways; researchers modulate neuronal activity, secretase function, or vesicle trafficking to control its release. This approach helps clarify activity-dependent Aβ signaling, peptide accumulation, synaptic dysfunction, and the conditions that promote aggregation, while supporting evaluation of therapeutics that target amyloid processing or secretion.

Fabrication of Amyloid-Beta-Releasing Polymer Microbeads for Modeling Alzheimer's Disease

This video demonstrates a method for fabricating polymer microbeads that encapsulate neurotoxic amyloid-beta-secreting cells. The cell-polymer suspension is processed through a microbead fabrication device, with polymer gelation achieved via calcium cross-linking, forming spherical microbeads around the cells. The polymer allows cell proliferation while allowing controlled, long-term amyloid-beta release, simulating its accumulation in Alzheimer's disease.

We present a facile method to fabricate a biodegradable gelatin-based drug release platform that is magneto-thermally responsive. This was achieved by incorporating superparamagnetic iron oxide nanoparticles and poly(N-isopropylacrylamide-co-acrylamide) within a spherical gelatin micro-network crosslinked by genipin, in conjunction with an alternating magnetic field application system.

We present a novel method that uses photo-responsive block copolymers for more efficient spatiotemporal control of gene silencing with no detectable off-target effects. Additionally, changes in gene expression can be predicted using straightforward siRNA release assays and simple kinetic modeling.
