Lidocaine Without Epinephrine

Lidocaine without epinephrine is a local anesthetic formulation that produces temporary, localized loss of sensation without the vasoconstrictor epinephrine, making it an important option in pharmacology and clinical care. Lidocaine works primarily by blocking voltage-gated sodium channels in nerve membranes, preventing sodium influx and the generation and propagation of action potentials; without epinephrine, it does not reduce local blood flow or slow anesthetic absorption through vasoconstriction. This formulation is used for infiltration anesthesia, minor procedures, and situations in which epinephrine is undesirable, while its comparatively shorter duration and potentially greater systemic absorption must be considered when selecting an anesthetic.

Knowledge of molecular mechanisms underlying gustatory transduction has recently enjoyed significant advances, largely due to using animal models. However, the wide diversity in taste sensitivity and specificity among mammals warrants studies in human tissue. We describe a biopsy technique to collect living taste cells from the papillae on human tongue.

This video and protocol demonstrate how to implant a glass-covered cranial window in rodents. These preparations can be used for chronic in vivo two-photon imaging of the neocortex over time scales of months. It may also be used for other types of imaging, including optical intrinsic signal imaging.
