Membrane Lipids

Três tipos de lipídios estão presentes na membrana – fosfoglicerídeos, esfingolipídios e esteróis.
Os fosfoglicerídeos, também conhecidos como glicerofosfolipídios, contêm uma estrutura de glicerol com um grupo fosfato hidrofílico e duas cadeias de acila graxa hidrofóbica, geralmente com 16 a 22 átomos de carbono de comprimento.
Alguns dos fosfoglicerídeos mais comuns presentes na membrana celular são fosfatidilcolina, fosfatidiletanolamina, fosfatidilserina e fosfatidilinositol - formados pela ligação de colina, etanolamina, serina e inositol ao grupo fosfato.
Os esfingolipídios são lipídios que contêm uma estrutura de esfingosina. A esfingosina é um aminoálcool com uma cadeia de hidrocarbonetos longa e insaturada. 
O esfingolipídeo mais simples é a ceramida, formada quando uma molécula de ácido graxo se liga ao grupo amino da esfingosina.
O grupo hidroxila restante da ceramida pode formar ésteres com outros grupos químicos, como fosfocolina ou carboidratos, formando esfingomielina ou glicolipídios, respectivamente.
O terceiro tipo de lipídios de membrana, os esteróis, como o colesterol, não podem formar independentemente uma bicamada lipídica. Em vez disso, eles se intercalam dentro de bicamadas fosfolipídicas e atuam como um tampão de fluidez para a membrana.

Os glicolipídios compreendem uma classe fisiologicamente importante dos lipídios da membrana que estão sempre presentes no lado externo, ou extracelular, da membrana. Nos eucariotos, eles são encontrados tanto na membrana plasmática quanto nas membranas intracelulares. O galactocerebrosídio, um glicolipídio simples, existe abundantemente nas bainhas de mielina junto com a esfingomielina. Os gangliosídios, os glicolipídios mais complexos, têm carga negativa devido à presença de ácido siálico e podem constituir até 10% do total de lipídios nas membranas das células nervosas. Membrane lipids give biological membranes their structure and help set their properties. In mammalian cells, lipids make up about 50% of the membrane on average. The amount can be much lower in the inner mitochondrial membrane or much higher in the myelin sheath around nerve cells.

Several phospholipids are common in mammalian membranes. These include phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, and sphingomyelin. At physiological pH, phosphatidylserine carries a negative charge, while the other three are neutral.

The fatty acids in membrane lipids can differ in carbon length and in saturation. Some are fully saturated, which means they have no double bonds. Others are monounsaturated with one double bond, or polyunsaturated with multiple double bonds.

Glycolipids are an important membrane lipid class that stays on the outer, or extracellular, side of the membrane. In eukaryotes, they are found in both the plasma membrane and intracellular membranes. Galactocerebroside is a simple glycolipid found in large amounts in myelin sheaths along with sphingomyelin.

Gangliosides are the most complex glycolipids. They are negatively charged because they contain sialic acid, and they can make up as much as 10% of the total lipids in nerve cell membranes. Their roles are also studied in viral and genetic diseases. GM1 acts as a cell surface receptor for cholera toxin, and excess GM2 ganglioside is seen in Tay-Sachs disease.

Sterols are another major class of membrane lipids. Plant-specific sterols can make up 30% to 50% of the total lipids in the plant plasma membrane. In eukaryotic membranes, cholesterol is also common and can account for almost 50% of the total lipids. Cholesterol in human cell membranes is slightly different from ergosterol, which is a common lipid in fungal membranes.
