A junção aderente é uma junção de ancoragem que mantém as células unidas em tecidos como o epitélio.
As junções aderentes são grandes complexos multiproteicos que compreendem três classes principais de proteínas - as caderinas adesivas, o citoesqueleto de actina de ancoragem e as cateninas ligantes.
A montagem da junção aderente começa com a polimerização dos monômeros de actina próximos à membrana celular para formar saliências chamadas lamellipodia.
Quando os lamelipódios das células vizinhas estão próximos o suficiente, as caderinas em sua superfície interagem e desencadeiam a localização de mais caderinas, formando aglomerados.
Esses aglomerados são estabilizados por cateninas beta, alfa e p120, que ajudam a recrutar mais complexos caderina-catenina para o local de crescimento.
À medida que a zona de contato entre as duas células se expande, as alfa-cateninas mudam a polimerização da actina de ramificada para não ramificada e as organizam paralelamente à membrana celular.
Esses filamentos de actina então se associam à miosina para formar feixes contráteis, formando assim uma junção aderente madura.
A junção liga o citoesqueleto de uma célula a outra, mantida unida por meio das interações extracelulares das moléculas de caderina.

Adherens Junctions

Fortes pontos de contato entre células adjacentes ancoram-nas umas às outras, formando tecidos. Essas junções de ancoragem são de dois tipos – junções aderentes e desmossomos. As junções aderentes são abundantes em tecidos como epitélio e endotélio, formando uma zona contínua de adesão chamada cinturão de adesão. Em outros tecidos, como o músculo cardíaco, eles aparecem como aglomerados, ligando as células para produzir uma contração coordenada do músculo cardíaco. As junções aderentes são dinâmicas As células endoteliais que revestem os vasos sanguíneos estão fortemente ligadas, formando uma barreira que impede a fuga de outras células e moléculas da corrente sanguínea para o tecido circundante. Porém, durante uma resposta imune, os leucócitos da corrente sanguínea devem passar por essa barreira e atingir o tecido alvo. Moléculas inflamatórias, como histaminas, e proteínas sinalizadoras, como o fator de crescimento endotelial vascular (VGEF), desencadeiam processos downstream, como fosforilação e internalização de caderinas presentes na superfície da célula endotelial. Isso leva à desmontagem das junções aderentes e aumenta a permeabilidade endotelial, criando lacunas que permitem a passagem de células imunológicas e outras moléculas. A manutenção e a quebra das junções aderentes são, portanto, reguladas dinamicamente em resposta a estímulos específicos. Junções aderentes e infecções Agentes infecciosos como o Helicobacter pylori podem entrar no tecido do hospedeiro rompendo as junções aderentes e movendo-se através das lacunas intercelulares. Essas bactérias produzem proteases que clivam o domínio extracelular das caderinas, causando assim a desmontagem da junção. As caderinas da superfície celular também são sequestradas como receptores por bactérias como o Streptococcus pneumoniae para aderir ao epitélio respiratório superior. No entanto, o sequestro mais impressionante talvez seja demonstrado pela Listeria monocytogenes. Esta bactéria expressa proteínas da parede celular que ligam as E-caderinas e desencadeiam o recrutamento de ɑ, β e p120-cateninas para o local. Esse recrutamento inicia a polimerização dinâmica da actina, formando saliências de membrana que envolvem a bactéria em um processo denominado fagocitose induzida. Adherens junctions are strong cell-to-cell anchors that help tissues stay connected. These anchoring junctions work with desmosomes, another type of junction that holds adjacent cells together. In many epithelial and endothelial tissues, adherens junctions form a continuous adhesion belt. In heart muscle, they often appear as clusters that help cells work together for coordinated contraction.

Adherens junctions are also dynamic, especially in the cells that line blood vessels. These endothelial cells form a barrier that normally keeps cells and molecules inside the bloodstream. During an immune response, leukocytes, or white blood cells, must cross this barrier to reach damaged tissue. Inflammatory molecules such as histamines and signaling proteins such as vascular endothelial growth factor, or VEGF, can trigger phosphorylation and internalization of cadherins on the endothelial cell surface. Cadherins are adhesion proteins that help cells stick together. When this happens, adherens junctions break down, permeability increases, and gaps open for immune cells and other molecules to pass through.

Some infections also take advantage of adherens junctions. Helicobacter pylori can disrupt these junctions by producing proteases that cut the extracellular part of cadherins. This breaks the junctions and helps the bacteria move through the spaces between cells. Other bacteria, such as Streptococcus pneumoniae, use cadherins as receptors to attach to the upper respiratory epithelium. Listeria monocytogenes uses a more complex strategy. It binds E-cadherins and recruits alpha-, beta-, and p120-catenins, which starts actin polymerization. This creates membrane protrusions that engulf the bacteria in induced phagocytosis, allowing it to enter a nonphagocytic cell and multiply inside the host.
