Separation of Sister Chromatids

No final da metáfase, os cromossomos biorientados se alinham na placa metafásica. Durante esta fase, os complexos de anéis de proteína coesina mantêm as cromátides irmãs juntas na região do centrômero e evitam que sejam separadas.
A progressão da metáfase para a anáfase é desencadeada pela fosforilação induzida por ciclina-Cdk de uma multissubunidade, a enzima ubiquitina ligase - o complexo promotor da anáfase, também conhecido como ciclossomo ou APC / C. A APC / C fosforilada se liga a uma proteína, Cdc20, formando um complexo ativo.
O complexo APC/C ativo reconhece uma proteína inibitória chamada securina que está ligada a uma enzima protease chamada separase. Antes desse reconhecimento, a securina inibe a atividade de separase.
O complexo APC/C ativo marca a securina com a proteína ubiquitina, direcionando-a para a degradação proteassômica. Destruição de liberações de securina separase.
O complexo ciclina-Cdk também regula negativamente a atividade de separase não ligada por meio da fosforilação inibitória.
O complexo APC/C ativo causa a ubiquitinação das ciclinas, direcionando-as para a degradação proteassômica. A destruição da ciclina remove a atividade enzimática das quinases dependentes de ciclina ou Cdks.
A inativação de Cdks permite que as enzimas fosfatase desfosforilem a separação e previne a refosforilação inibitória mediada por Cdk da separase. A desfosforilação permite que a enzima separase clive o complexo de anéis de coesina que mantém as cromátides irmãs unidas.
A clivagem da coesina resulta na perda absoluta da coesão da cromátide irmã, marcando a transição da metáfase para a anáfase. A perda de coesão permite que as cromátides irmãs se separem e se movam para pólos opostos do fuso.

Na transição da prófase para a metáfase, ocorre uma redução na coesão ao longo dos braços cromossômicos, resultando na resolução das cromátides irmãs. No entanto, as conexões residuais de coesina permanecem para manter as cromátides irmãs unidas até a transição da metáfase para a anáfase. A conexão residual evita qualquer separação prematura das cromátides irmãs, bloqueando os riscos de aneuploidia nas células-filhas. No início da anáfase, a separase, uma enzima proteolítica, é ativada. A separase ativada cliva a subunidade Scc1 dos anéis de coesina residuais, resultando em uma perda total de coesão. As cromátides-irmãs se separam na ausência de força para mantê-las unidas. A falta de coesão permite que as forças em direção aos pólos, exercendo força ao longo dos microtúbulos, puxem as cromátides separadas em direção aos pólos do fuso. Somente quando cada cromossomo se alinha corretamente no fuso mitótico, o SAC é desativado para permitir a fosforilação e ativação das subunidades reguladoras APC/C - CDC20 e CDH1. O CDC20 é fosforilado por Cdk1/ciclina B para formar APC/C-CDC20 ativo. O APC/C-CDC20 ativo catalisa então a degradação da securina, ciclina A e ciclina B para promover a transição anáfase. Como a atividade da Cdk é dependente da ciclina, a degradação da ciclina B resulta na perda da atividade da Cdk1. A perda do complexo Cdk1/ciclina B inativa o APC/C-CDC20, mas ativa outra subunidade regulatória, APC/C-CDH1, que sinaliza a conclusão da transição metáfase para anáfase. O APC/C-CDH1 ativo facilita a saída mitótica e estabiliza a fase G1 subsequente, evitando o acúmulo prematuro de ciclinas mitóticas. A mudança nas atividades da APC/C-CDC20 e da APC/C-CDH1 tem duas consequências significativas. Primeiro, essas subunidades regulatórias desencadeiam especificidades de substrato sobrepostas, porém distintas, e assim promovem a transição ordenada do ciclo celular. Em segundo lugar, o CDC20 e o CDH1 são regulados através de mecanismos diferentes. Quando o APC/C-CDC20 está ativo, o CDH1 sofre fosforilação inibitória pela ciclina B/Cdk1, impedindo-o de se ligar ao APC/C. Em contraste, a atividade do CDC20 é inibida por um complexo de checkpoint mitótico - uma multiproteína (BUBR1, BUB3, CDC20 e MAD2) ativada pelo SAC.
