Tecidos como a pele e o epitélio intestinal estão constantemente experimentando forças mecânicas externas, como beliscar ou esticar.
As junções aderentes que ancoram essas células epiteliais juntas podem mudar dinamicamente em resposta a essa tensão mecânica. Essa conversão de estímulos mecânicos em mudanças bioquímicas é chamada de mecanotransdução.
Nessas células, as junções aderentes formam uma zona contínua de fixação - o cinturão de adesão - compreendendo grandes aglomerados de caderinas, cateninas e feixes contráteis de actomiosina.
As alfa-cateninas nessas junções atuam como proteínas mecanossensoriais que mudam de conformação em resposta à tensão mecânica.
Quando o aumento das forças contráteis puxa os filamentos de actina, a alfa-catenina muda da conformação dobrada para a desdobrada.
Esse desdobramento abre um sítio de ligação críptico à vinculina, uma proteína de ligação ao citoesqueleto que permanece em sua forma fechada e inativa no citoplasma.
Ao se ligar à alfa-catenina, a vinculina muda a conformação para sua forma aberta e ativa.
A vinculina ativa promove o recrutamento de filamentos de actina adicionais para o local, fortalecendo assim a junção e distribuindo a força pelas células.

Tension Response at Adherens Junctions

As junções aderentes que ancoram as células são complexos multiproteicos que se adaptam dinamicamente a estímulos mecânicos, como forças de tração e tensão de cisalhamento. As proteínas mecanossensoriais nestas junções podem detectar tais estímulos mecânicos e sofrer uma mudança na sua conformação, resultando numa função alterada – um processo chamado mecanotransdução. α-Catenina como Proteína Mecanossensorial A α-catenina das junções aderentes é uma proteína alostérica com três domínios VH (homologia da vinculina) que pode se ligar a várias proteínas de ligação à actina, como vinculina, afadina e α-actinina. Em condições normais, a α-catenina é dobrada e estes domínios de ligação a VH são inacessíveis. O terminal N está ancorado à β-catenina e a extremidade C-terminal está ligada ao citoesqueleto da actomiosina. Quando a força de tração interna aumenta e a miosina II puxa o filamento de actina, a α-catenina ligada se desdobra e seus domínios VH ficam disponíveis para ligação. Papel da Vinculina na Mecanotransdução A vinculina é outra proteína dinâmica com um domínio de cabeça e cauda unidos por uma região de pescoço flexível. Na sua forma inativa, os domínios cabeça e cauda interagem entre si, impedindo a ligação de outras proteínas. No entanto, quando o domínio VH exposto da α-catenina se liga ao domínio da cabeça da vinculina, a proteína muda para uma conformação aberta. O domínio da cauda livre agora pode ligar-se à actina e recrutar mais filamentos de actina para esta junção. A região do pescoço aberto também se liga a várias proteínas que regulam a polimerização da actina, como a Arp2/3, reorganizando assim dinamicamente o citoesqueleto de actina em resposta ao estresse mecânico.

Adherens junctions help cells stay attached while they respond to mechanical force. These multi-protein complexes can adapt to tensile force and shear stress. When the force changes, mechanosensory proteins in the junction can change shape. This process is called mechanotransduction.

Alpha-catenin is one of the main force-sensing proteins in adherens junctions. It is an allosteric protein, which means its shape affects how it works. Alpha-catenin has three VH, or vinculin homology, domains that can bind actin-binding proteins such as vinculin, afadin, and alpha-actinin. Under normal conditions, alpha-catenin is folded, so these VH binding domains stay hidden.

The N-terminal end of alpha-catenin is attached to beta-catenin. Its C-terminal end is linked to the actomyosin cytoskeleton. When internal tensile force increases, myosin II pulls on the actin filament. This pull makes alpha-catenin unfold. Once it opens, its VH domains become available for binding.

Vinculin also helps the junction respond to force. It has a head domain and a tail domain joined by a flexible neck region. In its inactive form, the head and tail interact with each other, which blocks other proteins from binding. When the exposed VH domain of alpha-catenin binds the vinculin head domain, vinculin changes into an open shape.

In the open form, vinculin can use its tail domain to bind actin and recruit more actin filaments to the junction. The open neck region can also bind proteins that control actin polymerization, such as Arp2/3. Together, these changes help reorganize the actin cytoskeleton in response to mechanical stress.
