A coloração em microscopia aumenta a visibilidade dos microrganismos, distinguindo suas estruturas internas e externas. A coloração simples usa um único corante para destacar o tamanho, a forma e as características básicas de uma célula. A fixação é uma etapa crucial que preserva a morfologia celular e imobiliza as estruturas antes da coloração. A fixação por calor é comumente usada para amostras bacterianas e archaea em microscopia de luz. Para realizar uma coloração simples, um esfregaço fino de cultura microbiana é espalhado em uma lâmina, seco ao ar e fixado ao calor. A lâmina é então corada com um corante básico - como violeta cristal, azul de metileno ou safranina - por 1 a 2 minutos. Após a coloração, a lâmina é enxaguada com água para remover o excesso de tinta e seca com papel absorvente. Simple Staining Technique

Simple staining helps reveal the size, shape, and arrangement of microorganisms under the microscope. It uses one dye to color the whole cell. That creates strong contrast between the cells and the background.

A careful preparation step comes first. A small drop of microbial culture is spread on a clean glass slide to make a thin smear. The smear is air-dried and then heat-fixed, which means it is briefly passed over a flame.

Heat fixation keeps the cells attached to the slide. It also helps prevent autolysis, or self-destruction of the cells, and preserves their shape. In addition, it can improve dye uptake by changing the permeability of the cell wall.

After fixation, the slide is covered with a basic dye such as crystal violet, methylene blue, or safranin for 1 to 2 minutes. These dyes have positively charged chromophores that bind to negatively charged parts of the cell, including nucleic acids and the cell wall. The slide is then rinsed with water and blotted dry with absorbent paper.

The result is brightly stained cells on a clear background. This makes it easier to observe microbial morphology and arrangement. Simple staining does not show detailed structures such as organelles, and it does not tell Gram-positive and Gram-negative bacteria apart. Even so, it is a useful first step in microbiology and often comes before more advanced staining methods.
