The DNA Helix

Каждая из цепочек ДНК состоит из подразделений, называемых нуклеотидами, которые содержат сахар дезоксиридобозу, фосфатную группу, и одно из четырех азотсодержащих оснований: аденин (A), гуанин (G), цитозин (C) и тимин (T). Цитозин и тимин принадлежат к группе однокольцовых структур, называемых пиримидинами.

Соседние нуклеотиды в одной цепочке ковалентно связаны фосфодитерными связями. Две цепочки нуклеотидов содержатся вместе водородными связями, в которых аденины в одной цепочке пары с тиминами в том же положении в другой цепочке, и цитозины в одной нити пары с гуанинами в том же положении в другой нити. Это склеивание водорода является возможным благодаря антипараллельной компоновки двух цепочек ДНК, в которых 5' и 3' концы нитей ориентированы в противоположных направлениях. Без этого нуклеотидов было бы в неправильном положении, чтобы сформировать водородные связи между цепочками.

 Две цепи молекулы ДНК плотно закручены в пружинную структуру, называемую двойной спиралью. Однако двойная спираль не является идеально симметричной. <h4> Открытие структуры ДНК: краткая история </h4>

 История открытия структуры ДНК начинается в 1869 году, когда швейцарский ученый Фридрих Мишер открыл вещество, которое он назвал «нуклеином». В процессе извлечения белка из белых кровяных телец Мишер обнаружил неожиданное вещество с относительно высоким содержанием фосфора. Он не знал, что это было, но подозревал, что это может быть биологически важно. Мишер был прав, но научному сообществу потребовались десятилетия, чтобы полностью оценить его идеи. 

 Следующее критическое открытие сделал российский биохимик Фебус Левене. В 1919 году Левен предположил, что нуклеин, к тому времени известный как нуклеиновая кислота, состоит из цепочек молекул, которые он назвал полинуклеотидами. В начале 1950-х годов американский биолог Джеймс Уотсон и английский физик Фрэнсис Крик мчались со своим главным соперником, американцем Лайнусом Полингом, чтобы обнаружить трехмерную структуру ДНК.
