The Phragmoplast

Растительные клетки окружены жесткой клеточной стенкой, что затрудняет деление клеток путем сокращения извне. Следовательно, растительные клетки делятся, образуя новую клеточную стенку между двумя дочерними ядрами «изнутри наружу». Специализированная структура растений, называемая фрагмопластом, руководит формированием новой клеточной стенки.
Плоскость клеточного деления, которая является будущим местом формирования клеточной стенки, отмечена препрофазной полосой, состоящей из микротрубочек и актиновых филаментов, которая образуется во время G1-фазы клеточного цикла. Препрофазная полоса собирается в коре клетки и исчезает в начале М-фазы.
Во время митоза хромосомы разделяются и начинают перемещаться к противоположным полюсам. На полюсах вокруг разделенных хромосом образуется ядерная оболочка, и веретена начинают исчезать. Остальные микротрубочки митотического веретена образуют фрагмопласт.
Микротрубочки фрагмопластов полярные, с плюсовыми концами вблизи экваториальной плоскости и минусовыми концами вблизи полюсов.
Тельца Гольджи выделяют небольшие везикулы, которые разносятся моторными белками по микротрубочкам к центру клетки. Эти везикулы наполнены полисахаридами и гликопротеинами, двумя важными компонентами, необходимыми для синтеза новой клеточной стенки. Слияние везикул образует дискообразную структуру, называемую ранней клеточной пластиной.
Микротрубочки фрагмопластов успешно регенерируются на свободных краях клеточной пластины, что позволяет фрагмопласту удлинять пластинку в латеральном направлении. Новая клеточная пластина соединяется со стенками материнской клетки в месте, которое было отмечено препрофазной полосой до М-фазы. Фрагмопласт

Деление клеток необходимо для роста и развития организма. В клетках животных центральное веретено и связанные с ним белки образуют среднее тело — структуру, которая играет важную роль в цитокинезе. У растений центральное веретено вместе с микротрубочками, актином и другими компонентами клетки созревает во фрагмопласт, необходимый для цитокинеза. В отличие от неподвижного среднего тела, фрагмопласт расширяется центробежно, что в конечном итоге приводит к образованию новой клеточной стенки. Зрелый фрагмопласт имеет форму бублика и содержит внешнюю ведущую область, среднюю переходную зону и внутреннюю отстающую область. В ведущем регионе непрерывно собираются новые микротрубочки. Зарождение микротрубочек инициируется кольцевым комплексом γ-тубулина (γ-TuRC), который содержит шесть субъединиц — от GCP1 до GCP6. Однако γ-TuRC не может напрямую взаимодействовать с микротрубочками; следовательно, augmin, белковый комплекс, взаимодействующий с микротрубочками, опосредует рекрутирование γ-TuRC в микротрубочки. Микротрубочки, присутствующие во фрагмопласте, действуют как пути для транспортировки секреторных пузырьков, вырабатываемых близлежащим тельцем Гольджи. В средней переходной зоне везикулы сначала переходят в канальцевые структуры, а затем соединяются между собой, образуя переплетенную канальцевую сеть. Непрерывное добавление секреторных пузырьков приводит к росту канальцевой сети наружу до тех пор, пока она не встретится с мембраной родительской клетки. Мембраны везикул используются для формирования плазматической мембраны дочерних клеток, а биомолекулы, присутствующие в везикулах, используются для построения клеточной пластинки, расположенной между мембранами двух дочерних клеток. В отстающей области микротрубочки деполимеризуются, оставляя после себя зрелую часть клеточной пластинки. Полностью сформированная клеточная пластинка впоследствии превращается в клеточную стенку, разделяющую две дочерние клетки. Полимеризация микротрубочек на переднем конце и деполимеризация микротрубочек на отстающем участке вызывают расширение фрагмопласта и клеточной пластинки наружу до тех пор, пока последняя не соединится с материнской клеточной стенкой. Нарушение функций фрагмопластов может привести к образованию многоядерных клеток и эмбриональным летальным фенотипам.
