Replication in Eukaryotes

Репликация ДНК в эукариотических хромосомах начинается в нескольких источниках репликации, которые идентифицируются и связываются с помощью комплекса распознавания происхождения, или ORC.

Репликация у эукариот

В эукариотических клетках репликация ДНК высококонсервативна и строго регулируется. Перед делением клетки необходимо дублировать несколько линейных хромосом с высокой точностью, поэтому существует множество белков, которые выполняют специализированные роли в процессе репликации. Репликация происходит в три фазы: инициация, элонгация и терминация, и заканчивается наличием в ядре двух полных наборов хромосом. 

 Многие белки управляют репликацией в самом начале 

 Репликация эукариот следует во многом тем же принципам, что и репликация ДНК прокариот, но поскольку геном намного больше, а хромосомы имеют линейную, а не кольцевую форму, этот процесс требует большего количества белков и имеет несколько ключевых отличий. Во-первых, в отличие от прокариот, репликация у эукариот происходит одновременно в нескольких точках начала репликации вдоль каждой хромосомы. Белки-инициаторы распознают эти источники происхождения, связываются с ними и привлекают белки геликазы для раскручивания двойной спирали ДНК. В каждой точке происхождения образуются две репликационные вилки. Затем примаза добавляет короткие праймеры РНК к одиночным цепям ДНК, которые служат отправной точкой для связывания ДНК-полимеразы и начала копирования последовательности. ДНК может быть синтезирована только в направлении от 5' к 3', поэтому репликация обеих цепей из одной репликационной вилки происходит в двух разных направлениях. Ведущая цепь синтезируется непрерывно, а отстающая цепь синтезируется короткими участками длиной 100–200 пар оснований, называемыми фрагментами Оказаки. После завершения основной части репликации ферменты РНКазы удаляют праймеры РНК, ДНК-полимераза заполняет пробелы, а ДНК-лигаза запечатывает пробелы в новой цепи. 

 Разделение работы репликации между полимеразами 

 Рабочая нагрузка по копированию ДНК у эукариот разделена между несколькими различными типами ферментов ДНК-полимеразы. Основные семейства ДНК-полимераз во всех организмах классифицируются по сходству их белковых структур и аминокислотных последовательностей. Первые обнаруженные семейства были названы A, B, C и X, а семейства Y и D были идентифицированы позже. Полимеразы семейства B у эукариот включают Pol α, который также действует как примаза в репликационной вилке, а также Pol δ и ε, ферменты, которые выполняют большую часть работы по репликации ДНК на ведущей и отстающей цепях матрицы соответственно. Другие ДНК-полимеразы отвечают за такие задачи, как восстановление повреждений ДНК, копирование митохондриальной и пластидной ДНК, а также заполнение пробелов в последовательности ДНК на отстающей цепи после удаления праймеров РНК. 

 Теломеры защищают концы хромосом от деградации 

 Поскольку эукариотические хромосомы линейны, их концы подвержены деградации. Чтобы защитить важную генетическую информацию от повреждения, концы хромосом содержат множество некодирующих повторов высококонсервативной богатой G ДНК — теломер.
