Cell Detachment Control

Cell detachment control is the deliberate regulation of how and when adherent cells are released from culture surfaces or engineered biomaterials, a key requirement for reproducible bioengineering workflows. It works by modulating cell-substrate adhesion, including integrin-mediated interactions with extracellular matrix proteins, through biochemical treatments, temperature or surface cues and, when needed, applying controlled mechanical forces to limit damage and preserve viability. This control supports cell harvesting, passaging and downstream analysis, while enabling engineered tissues, cell-based assays and biomaterials with more consistent cell density, phenotype and function.

Creating experimental retinal detachments with a reproducible and sustained height of detachment, and without subretinal hemorrhage, is important for studying the pathophysiology of photoreceptor cell loss in retinal disease and evaluating potential therapeutic interventions. Here, we present a protocol for photoelectron imaging of anionic species. Anions generated in vacuo and separated by mass spectrometry are probed using velocity mapped photoelectron imaging, providing details of anion and neutral energy levels, anion and neutral structure and the nature of the anion electronic state.

This manuscript details an optimized inoculation protocol that uses detached maize leaf sheaths for reproducible cytological, physiological, and molecular studies of maize interactions with fungal plant pathogens. The leaf sheaths facilitate real-time observation of cellular interactions between the living plant and fungus in unfixed tissues.
