Human Tonsil Tissue

Human tonsil tissue is lymphoid tissue located at the entrance to the pharynx, where it contributes to immune surveillance of inhaled and ingested material. Its specialized epithelium exposes antigens to underlying antigen-presenting cells, while B and T lymphocytes coordinate germinal-center responses that support antibody production and immunological memory. In immunology and infection research, human tonsil tissue provides a clinically relevant model for studying mucosal immunity, host-pathogen interactions, lymphocyte activation, and responses to vaccination. Analyses of tonsil-derived cells and tissue architecture can clarify how local immune responses develop and may inform investigations of infectious disease and immune dysfunction.

Palatine tonsils are a rich source of B and T lymphocytes. Here we provide an easy, efficient and rapid protocol to isolate B and T lymphocytes from human palatine tonsils. The method described has been specifically adapted for studies of the viral etiology of tonsil inflammation known as tonsillitis.

Methods to create human 3D tumor tissues as test systems are described.
