Neutrophil Detection

Neutrophil detection comprises methods used to identify and quantify neutrophils in blood, tissues, or experimental samples, making it essential for studying innate immune responses and infection. Depending on the sample and research question, detection relies on cell morphology, surface markers such as CD66b or Ly6G, or lineage-associated proteins including myeloperoxidase, measured through microscopy, immunostaining, flow cytometry, or blood analysis. These approaches reveal neutrophil recruitment, activation, and accumulation during inflammation, infection, and tissue injury. Accurate detection supports diagnosis, immune profiling, evaluation of host-pathogen interactions, and assessment of therapies that alter inflammatory responses.

In this video, we demonstrate an assay utilizing fluorescence microscopy to detect the adhesion of cancer cells to neutrophil extracellular traps (NETs). In the test wells, there is a notable increase in the number of fluorescently labeled cancer cells adhering to NETs, whereas wells treated with DNase exhibit reduced cancer cell adhesion to NETs.

We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.
