Enzyme-linked Immunosorbent Assay

Enzyme-linked immunosorbent assay (ELISA) is a laboratory method that detects and measures specific antigens or antibodies in biological samples, making it important for medical diagnosis and research. The assay immobilizes a target or capture antibody on a solid surface, uses selective antigen–antibody binding, and links an enzyme to a detection antibody; adding a suitable substrate produces a measurable color change related to the amount of target present. ELISA supports testing for infectious diseases, immune responses, hormones, and other biomarkers, while its sensitivity, specificity, and scalable format make it valuable for clinical screening, disease monitoring, and biomedical investigation.

In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane. There are many different types of ELISAs, but they all involve an antibody molecule whose constant region binds an enzyme, leaving the variable region free to bind its specific antigen. Enzyme-substrate reaction allows the antigen to be visualized or quantified.
