Water Bath Sectioning

Water bath sectioning is a histological technique used to flatten and transfer thin sections of paraffin-embedded tissue onto microscope slides, reducing folds and wrinkles that can obscure cellular detail. After a microtome cuts the tissue block, sections are placed on a warm water surface, where controlled heat allows the paraffin ribbon to spread without melting the section; the flattened sections are then collected on coated slides and dried. In neuroscience, this preparation supports consistent visualization of brain and spinal cord architecture, neuronal morphology, lesions, and staining patterns, improving the quality and interpretability of microscopic analysis in research and teaching.

Here, we present a protocol to demonstrate the generation of ice when water is introduced to a cold bath of brine, as a secondary refrigerant, at a range of temperatures well below the freezing point of water. It can be used as an alternative way of producing ice for industry.

The present protocol describes the maceration and cleaning of cadaveric bone with a vacuum-sealed, hot water bath immersion technique. This is a low-cost, safe, and effective method to produce anatomical specimens for surgical planning and medical education as an alternative to three-dimensional (3D) printed models.
